Objective:To explore the effect of circadian clock gene polymorphisms on the occurrence of essential hypertension(EH)in the population from the perspective of population genetic susceptibility.Methods:This study employed a case-control research method,obtained general information of the research subjects through a structured questionnaire,and candidate genes and functional SNP sites were selected through the public database SNP info.Genotyping were performed on the iPLEX GOLD technology system(Sequenom)of the Sequenom MassARRAY molecular weight array technology platform.Pearson?~2test was used to compare the distribution of each SNP genotype in cases and controls.In addition,non-conditional logistic regression was used to analyze the relationship between candidate SNPs and the risk of EH.At the same time,potential confounders such as gender,age,smoking,and drinking were adjusted.and the adjusted OR and 95%CI of homozygotes,heterozygotes,dominant models,recessive models,and additive models were calculated.Results:Compared with wild homozygote(TT),the rs17060415(RORB,RAR-related orphan receptor B)locus increased the risk of EH(OR=1.7,95%CI:1.1,2.64).In the model,carrying the T site might reduce the risk of EH(OR=0.59,95%CI:0.38,0.9)comparing with the C site.In the recessive model,rs4795424(NR1D1,Nuclear receptor subfamily 1,group D,member 1)(A vs C,OR=0.75,95%CI:0.58,0.98),rs3027178(PER1,Period Circadian Regulator 1)(G vs T,OR=0.78,95%CI:0.61,0.99),rs4376556(RORB)(A vs G,OR=0.60,95%CI:0.41,0.86),rs7846903(RORB)(A vs G,OR=0.60,95%CI:0.41,0.87),rs9017(RORC,RAR-related orphan receptor C)(T vs C,OR=0.77,95%CI:0.60,0.99)loci were negatively correlated with EH,rs228729(PER3,Period Circadian Regulator 3)was positively correlated with EH(T vs C,OR=1.30,95%CI:1.02,1.65).In the additive model,rs4376556,rs7846903,and rs9017 were still negatively correlated with EH.Conclusion:The genetic variation of the RORB-rs17060415 locus increases the risk of EH.Per3-rs228729 was positively correlated with the risk of EH,while RORB-rs4376556,RORB-7846903,RORC-rs9017,NR1D1-rs4795424,and Per-rs3027178 were negatively associatedwith the risk of EH.
