MiR-495-3p Inhibits Proliferation And Migration Of Hepatoma Cell Line By Down-regulating SATB1 Expression | Posted on:2020-12-20 | Degree:Master | Type:Thesis | Country:China | Candidate:D Gong | Full Text:PDF | GTID:2404330629986220 | Subject:Pathology and pathophysiology | Abstract/Summary: | PDF Full Text Request | Background:MiRNA-495-3p is involved in the proliferation and migration of various cancer cells,and plays an important regulatory role in the function of cancer cells.Special at-rich binding protein-1(SATB1)is a protein that plays an important role in the growth and development of thymus and the growth and differentiation of T cells.It plays an important role in the proliferation,invasion and metastasis of tumors by regulating cell cycle,inducing cell differentiation and inhibiting apoptosis.Objective:This experiment mainly investigated the difference of miR-495-3p and SATB1 expression in normal liver cells and liver cancer cells.To determine whether miR-495-3p could inhibit the proliferation and migration of liver cancer cell line SMMC-7721 by regulating the expression of SATB1 protein.Methods:1.The expression level of miR-495-3p SATB1 mRNA in normal liver cells and liver cancer cell line SMMC-7721 was detected by real-time fluorescence quantitative PCR.The difference of SATB1 protein expression in normal liver cells and liver cancer cell line SMMC-7721 was detected by Western blot.2.The expression level of miR-495-3p in liver cancer cell line SMMC-7721 was up-regulated by cell transfection technology.Transwell assay was used to detect the migration ability of liver cancer cell lines SMMC-7721 in different treatment groups.3.The expression level of miR-495-3p in liver cancer cell line SMMC-7721 was up-regulated by cell transfection.The expression level of SATB1 protein and MMP-2 ?MMP-9 in liver cancer cell line SMMC-7721 in different treatment groups was detected by Western blot.4.The expression level of miR-475-3p in liver cancer cell line SMMC-7721 was up-regulated by cell transfection technology.And the expression level of SATB1 overexpression plasmid in the same group of liver cancer cell line SMMC-7721 was detected by Western blot method.Results:1.The expression level of miR-495-3p in liver cancer cell line SMMC-7721 was significantly lower than that in normal liver cells,while the expression level of SATB1 mRNA and protein was significantly higher than that in normal liver cells.2.The up-regulated expression of miR-495-3p in SMMC-7721 could inhibit the proliferation and migration of HCC cell line SMMC-7721.3.The upregulated expression of miR-495-3p in the cell line SMMC-7721 could significantly down-regulate the expression of SATB1 and MMP-2 ?MMP-9,the transport-related protein.4.The up-regulation of miR-495-3p expression level in cell line SMMC-7721 and the overexpression of SATB1 protein could significantly inhibit the regulation of miR-495-3p on the expression of MMP-2 ?MMP-9,a protein related to cell migration.Conclusion:1.MiR-495-3p was low expressed in liver cancer cell line SMMC-7721,while SATB1 was high expressed in liver cancer cell line SMMC-7721.2.MiR-495-3p can inhibit the proliferation and migration of liver cancer cell line SMMC-7721 and down-regulate the expression of SATB1 MMP-2 and MMP-9 proteins.3.MiR-495-3p inhibits the migration of HCC cell line SMMC-7721 by regulating SATB1 protein expression. | Keywords/Search Tags: | miR-495-3p, SATB1, HCC cells, proliferation, migration | PDF Full Text Request | Related items |
