Background:Metabolic syndrome seriously threatens human health and brings a heavy economic burden to society.Excessive intake of fructose can induce metabolic syndrome and increase the risk of chronic diseases such as renal injury.Conjugated linoleic acid(CLA),a nutrient element,has a number of physiological activities such as improving insulin resistance and anti-inflammatory.In this study,a fructose diet was used to build a rat model of metabolic syndrome,to explore the effect of CLA on the fructose-induced metabolic disorder and the protective effect of the kidney.Objective:The chief aim of our study was to investigate the effects and potential mechanisms of CLA on fructose-induced metabolic disorders and renal protection.Methods:Rats were randomly divided into normal group,model group and CLA group.Both model and CLA group were given 10% fructose drinking water for 6 weeks.At the same time,the rats of CLA group were administrated with CLA(200 mg/kg)by gavage every day.We measured the non-invasive blood pressure in rats.To evaluate renal function,we detected the levels of uric acid,creatinine and uric acid nitrogen.To evaluate insulin resistance,we measured fasting blood glucose,fasting insulin,and calculated HOMA-IR and QUICKI.HE staining was used to observe renal pathological changes.The levels of IL-1?,IL-18,TNF-? and IL-6 in serum and kidney were detected by ELISA.Western blot was used to detect the expression of NLRP3 inflammasome and NF-?B protein in the kidney.Results:In the fructose model group,the systolic blood pressure and diastolic blood pressure significantly increased.After oral administration of CLA,the systolic blood pressure significantly decreased.In the model group,the levels of serum uric acid and creatinine were significantly increased,and the level of urine uric acid was significantly reduced,while CLA intervention could reverse this situation.CLA could reduce fasting blood glucose and fasting insulin,and significantly improve fructose-induced insulin resistance.Renal pathological section HE staining showed that CLA supplement could reduce fructose-induced renal pathological injury.Moreover,CLA could significantly reduce the levels of inflammatory factors including IL-1?,IL-18,TNF-? and IL-6 in serum and kidney,and ameliorate renal inflammation.In addition,CLA decreased the release of inflammatory factors,and protected the kidney by inhibiting the activation of NLRP3 inflammasome and the NF-?B signaling pathway.Conclusion:CLA can significantly ameliorate fructose-induced metabolic disorders and renal injury.CLA may be to inhibite the NLRP3 inflammasome and NF-?B signaling pathway,decrease the release of inflammatory factors,relieve renal inflammation,and protect the kidney.
