Candida albicans,as the most common pathogenic fungus,lives in the human oral cavity,skin,respiratory tract,digestive tract,vagina and other mucous membranes in general.It can exist in three states of yeast,hyphae,and pseudohyphae,which can cause superficial mucosal and skin infections.It can also cause deep infections,which will be fatal.At present,with the increasing incidence of infection and mortality,Candida albicans is becoming more and more important as a pathogen.Therefore,the current research on the molecular pathway of the pathogenicity of Candida albicans is helpful for better research on new drugs or modification of existing drugs.Although there is no research on Zds1p in Candida albicans,it has been confirmed in other fungi that Zds1p plays an important role in bacterial cell life activities,such as cytokinesis and cell wall integrity.In this paper,the function of Zds1p in Candida albicans has been identified.When the Zds1p gene is knocked out or turned off in the strain,the growth vigor decreases obviously and pseudohyphae growth is obvious.The cytoplasmic division of zds1Λ/Λis damaged and multinucleated.And in the induction medium,the ability of the knockout strain to be induced to grow hyphae is defect,and the toxicity to the cells also decreased.Combined with transcriptome sequencing,it is found that after deletion of the ZDS1 gene,the expression of many key genes in the cell cycle pathway,the MAPK pathway,and meiosis pathways are down-regulated,indicating that Zds1p is involved in regulating the process of cytokinesis and cell cycle.In addition,Zds1p can participate in regulating the hypha growth of Candida albicans,which in turn affects the infectivity of the strain.The zds1 knockout strain is sensitive to zymolyase-20T,and it’s cell wall is thicker and more transparent than the wild-type strain,which indicates that the loss of Zds1p will affect the integrity of the cell wall.It is also more sensitive to calcium ions,with lower tolerance for drugs such as amphotericin B and nystatin,but higher tolerance for drugs such as fluconazole and ketoconazole than wild-type strains.Combined with transcriptome sequencing found that the expression of the gene ERG3decreased in the knockout strain,indicating that the Zds1p is involved in the stress response of drug in a certain way,and the specific role is in the further study.Taken together,Zds1p participates in multiple life activities in Candida albicans,including cell cycle,cell division,Ca2+tolerance,drug stress response,cell wall integrity,viability and cell morphology in culture media.The RHO family is a very important superfamily in Candida albicans,and Cdc55 PP2A as a new effector of Rho1 is also a very important serine/threonine kinase.Zds1p has been shown to play an important role along with Cdc55 in Saccharomyces cerevisiae,and can also interact with Rho1 in vivo.and In addition,the results of transcriptome sequencing in this subject has shown that ZDS1 can also regulate the expression of RHO1 and CDC55.Therefore,this project attempts to detect proteins that can directly interact with Zds1p by Western Blot in vitro.It is still in the preliminary experimental stage.
