ObjectiveTo investigate the effect of mesenchymal stem cell supernatant of different concentrations on the activity of echinococcus granulosus and the concentration difference.Methods1.Extraction and culture of MSCs from the lower limb long bones of C57 mice aged 4-6 weeks Morphological characteristics of MSCs were observed by microscopy;2.Observe the morphology and growth of MSCs under a microscope;3.MSCs were cultured,passaged,and the supernatant was taken and diluted to 100% and 50% concentration;4.The experimental groups were cultured using different concentrations of MSC supernatants to culture the protocephala,and the eosin staining was observed and counted at different time points,Save the specimen to detect the Caspase-3 enzyme activity and prepare the electron microscope specimen;5.Follow the steps of the Caspase-3 enzyme kit to detect the enzyme activity in the specimen,and observe the micromorphological changes of the protocephala at different time points under an electron microscope;6.Experimental data processing and statistical analysis by SPSS 17.0.Results1.Mesenchymal stem cell supernatant and fine-grained ganglion has improved its survival rate In vitro culture.2.The supernatant of mesenchymal stem cells reduced the activity of Caspase-3 enzyme in fine-grained ganglion in vitro.The Caspase-3 activity of the experimental group and the control group was statistically significant(P <0.05)3.Observation under the electron microscope of the protocephala experimental group was more complete than that of the control group,and the local structure was different.Conclusion The mesenchymal stem cell supernatant can promote the activity of E.granulosus protozoan activity in vitro.
