Objective:In this experiment,by extracting Berberine,an effective component of Coptis chinensis, the effects of berberine on body weight, blood glucose, blood lipids, liver weight, liver pathological changes, and endoplasmic reticulum stress signals were investigated in diabetic rats.Regulating effects on liver endoplasmic reticulum stress in diabetic rats.Methods:60 male SD rats were selected. After 7 days of adaptive feeding,10 rats were randomly selected as the experimental blank group and fed with normal feed, and the remaining 50 rats were given high-fat and high-sugar feed.Come feed. After feeding for 4 weeks by the above method, rats except the NC group will be fasted for 12 hours, and then intraperitoneal injection of streptozotocin(30 mg·kg-1,with pH 4.4 0.1 mol·L-1citrate buffer dissolved),and then continue to feed high-fat high-sugar feed.In the NC group,an equal amount of citrate buffer was injected intraperitoneally. After waiting for 72 hours, the fasting blood glucose after 12 hours of fasting was detected by tail-cutting blood collection. If FBG?16.7mmol·L-1detected for 3 consecutive days, the model of type 2 diabetic rats was successful If an unsuccessful model is encountered,an additional injection of STZ?12.5mg/kg?is given.Fifty diabetic rats were selected and randomly divided into model group, berberine low, medium and high dose groups, metformin group,10 rats in each group.Rats in the low, medium and high dose groups of berberine were gavaged at 75 mg,150 mg and 300 mg/kg,respectively;rats in the metformin group were gavaged at 200 mg/kg;Gavage was performed according to the same volume of normal saline.Once a day, continuous gavage for4 weeks. The rats will be tested for blood glucose and body weight once a week during gavage. After 4 weeks of intragastric administration, the rats were fasted for 16h,then intraperitoneal injection of pentobarbital to anesthetize, intubate the right common carotid artery to collect blood samples, remove the blood samples in a centrifuge at 4?,1500r/min The plasma was collected by centrifugation for 15 min under the conditions, and used in the later biochemical kit to detect blood glucose and blood lipid levels. The liver was quickly peeled off by deplaning, washed with normal saline, blotted with filter paper, and weighed with an electronic balance.Part of the liver tissue was taken for pathological section in each group. The remaining tissue was quickly placed in a refrigerator at-80°C for testing.Indicator GRP78,p-Perk,p-Eif2?expression levels.Results:?1?Effects of berberine on body weight:With the progress of the experiment, the body weight of the NC group gradually increased.Compared with the NC group, the body weight of the DM group showed a downward trend?P<0.01?.At 2 weeks, the weight of the rats in the MF group rebounded. At 3 and 4 weeks, there was significant statistical significance?P<0.01?.In the first two weeks, the changes in body weight of berberine at low, medium and high doses were Compared with the DM group, there was no statistical significance at the 3rd and 4th week. The body weight of the BH group was significantly higher than that of the DM group?P<0.01?.?2?Effect of berberine on fasting blood glucose:Compared with the NC group, the blood glucose level in the DM group was higher?P<0.01?,indicating successful modeling. At week 1,metformin and high-dose berberine can reduce fasting blood glucose in rats?P<0.05?.At week 2,the blood glucose levels of ME group and BM group decreased significantly?P<0.01?,at week 3 And in the 4th week, the blood glucose of ME group,BL group,BM group and BH group decreased significantly?P<0.01?.?3?Effect of berberine on liver weight:Compared with the NC group, the liver weight of the DM group increased significantly?P<0.01?.Compared with the DM group, the liver weight of the ME group,BM group, and BH group decreased significantly?P<0.01?.?4?Effect of berberine on blood lipids:Compared with the NC group, the levels of TC,TG,LDL-C in blood lipids increased, and the levels of HDL-C decreased?P<0.01?.Compared with the DM group, the TC,TG and LDL-C levels of the MF group showed a significant downward trend, while the HDL-C levels showed an upward trend?P<0.01?;the TC,LDL of the BM group-C level decreased?P<0.01?,TC,TG,LDL-C levels of BH group decreased significantly, and HDL-C level increased?P<0.01?.?5?Liver tissue oil red O staining and HE pathological results:The results of liver tissue oil red O staining and HE pathological section showed that berberine can significantly reduce the inflammatory cell infiltration and lipid deposition in liver tissue of DM rats,so it shows that berberine can be significantly reduced due to diabetes And cause liver damage.?6?Effect of berberine on the expression levels of GRP78,p-Perk and p-Eif2?:Studies have found that the expression of GRP78,p-Perk,and p-Eif2?related proteins of endoplasmic reticulum stress in liver tissues of diabetic rats Increased, while medium and high doses of berberine can down-regulate the expression of GRP78,p-Perk,and p-Eif2?,and medium doses have the best effect. Conclusion:Through this experiment, after 4 weeks of berberine treatment, compared with the model group, the body weight of diabetic rats increased, the level of fasting blood glucose decreased significantly, the levels of TC,TG,LDL in blood lipid decreased, and the level of HDL increased,Decreases the weight of the liver, and pathological slices show that it can significantly improve liver damage and show good protection for the liver. The mechanism may be through GRP78,p-Perk,and p-Eif2?endoplasmic reticulum stress signaling pathways.Effective.
