Background:Diffuse large B-cell lymphoma(DLBCL)is the most common non-Hodgkin B-cell lymphoma(B-NHL),and this kind of lymphoma is the most heterogeneous.The histomorphologic,immunophenotypic,and biologic behavior overlaps with Hodgkin lymphoma(HL),which is also a B cell source,making the clinicopathological diagnosis extremely difficult.OCT2 and BOB.1 are common transcription factors of B cells,which play an important role in the genesis and development of B cells and are normally expressed in germinal center and mantle lymphocytes of lymphoid tissue.Studies have shown that abnormalities of OCT2 or BOB.1 can promote the formation of B-cell lymphoma.Objective:To explore the expression patterns of OCT2 and BOB.1 in DLBCL and HL respectively,and to analyze and summarize the difference of expression between these two types of lymphoma.And the aim is to provide new ideas for the diagnosis and differential diagnosis of DLBCL and HL.Methods:1.40 cases of DLBCL and 35 cases of HL were collected from the Department of Pathology of the second affiliated Hospital of Nanchang University from January2016 to December 2018.2.All slices were stained with HE and immunohistochemical staining to detect the expression of OCT2 and BOB.1 in the two groups of patients.At the same time,the markers such as CD20,PAX5 and CD30 were detected.Combined with these markers,the slices of the two groups were read comprehensively.3.Statistical methods: The SPSS17.0 version of the statistical software was used,the T test was used for the measurement data,and the Chi-square test was used for the counting data,and the test level was ?= 0.05(bilateral).Results:1.There was significant difference in the age of onset of lymphoma types between DLBCL and HL groups(P < 0.05),no significant difference in sex and primary site(P > 0.05),and no significant difference in the maximum diameter of enlarged lymph nodes,with or without B symptoms and Ann-Arbor stage(P > 0.05).2.There was significant difference in the expression of OCT2,CD20 and CD30 between DLBCL and HL groups(P < 0.05),but there was no significant difference in the expression of BOB.1 and PAX5 between the two groups(P > 0.05).(1)OCT2expression: of the 40 patients DLBCL,39(97.5%)showed strong and diffusively positive OCT2 expression,which was expressed on the nucleus,and 1(2.5%)showed weak OCT2 expression.Among the 35 cases of HL,11(31.4%)of the R-S cells had moderately to moderately strong nuclear expression,and 24(68.6%)did not.(2)BOB.1 expression: Among the 40 DLBCL patients,27(67.5%)had positive expression of BOB.1,Pattern(1): 6 cases(15%)had weak positive expression of large cells,and the positive expression was located in the nucleus,and the whole was mottled.Pattern(2): 21 cases(52.5%),diffuse and uniform large cell nuclei were moderately positive,among which there were also scattered large cells with strong positive nuclei.13 cases(32.5%)had no expression.Among the 35 HL patients,16cases(45.7%)had positive expression of BOB.1.Pattern(1): 5 cases(14.3%)had very weak positive expression of R-S cells,which were expressed in the nucleus and discontinuous.Pattern(2): 8 cases(22.8%)were weakly positive and discontinuous in the nucleus.Pattern(3): 3 cases(8.6%)showed moderately strong and continuously expressed in the nuclear membrane.19 cases(54.3%)were silent.Conclusion:1.The positive expressions of OCT2 and BOB.1 in DLBCL were stable,OCT2 was uniformly strong positive,and BOB.1 was uniformly weak or moderate positive.2.The positive expression patterns of OCT2 and BOB.1 in HL were diverse,ranging from weak to moderate intensity.OCT2 was localized in the nucleus,while BOB.1 was localized in the nuclear membrane.3.Differences in the expression patterns of OCT2 and BOB.1 in DLBCL and HL,combined with other immune markers(such as CD20,PAX5,CD30,etc.),as well as clinical information and morphological characteristics,can be used as important indicators for the diagnosis and differential diagnosis of DLBCL and HL.
