Objective:1.To explore the established method and identification of patient-derived xenograft models of the prostate squamous cell carcinoma.2.The clinical cisplatin combined with mitoxantrone chemotherapy is used to verify whether the patient-derived xenograft models of prostate squamous cell carcinoma has a role of tumor inhibition,and it is initially confirmed that patient-derived xenograft preclinical model is successfully established.3.Initially verifying whether patient-derived xenograft models of the prostate squamous cell carcinoma can be used as a research tool for preclinical new drug screening.Methods:The treated surgical specimens of prostate squamous cell carcinoma were transplanted subcutaneously under the scapular region of Balb/c-Nu nude mice using a inoculating needles,and the xenograft tumor model was established and stably passaged.Then the homology between the xenograft tumor tissue and the original tumor after passage was preliminarily detected by histopathological HE staining and morphological immunohistochemical molecular method,The P3 generation tumor was inoculated subcutaneously into Balb/c-Nu nude mice,and when the tumor volume was 100-200mm3,the combination of cisplatin and mitoxantrone chemotherapy,which had been clinically used,was selected for drug validation.By regularly measuring the tumor length(L),width(W),and calculating the tumor volume,based on the experimental results of the two groups to initially verify whether the construction of prostate squamous cell carcinoma PDX model success.Results:The pathological HE staining morphological and immunohistochemicalmolecular expressions of xenograft model samples of the P3 generation nude mice are basically consistent with the original tumor;compared with the control group after injection of drugs,the value of measured volume-average combined the cisplatin and mitoxantrone group is significantly lower than that of the control group,and there is a significant difference between the two datas(P <0.05),indicating that the patient-derived xenograft model of prostate squamous cell carcinoma shows a certain inhibitory effect on the progression of prostate squamous cell carcinoma,which is basically consistent with the known reports.Conclusion:The patient-derived xenograft model of prostate squamous cell carcinoma is successfully established and the xenograft is stable.The combination of cisplatin and mitoxantrone was found to be effective in inhibiting tumor growth through the tumor growth inhibiting rate.Therefore,it is preliminarily concluded that the PDX model of prostate squamous cell carcinoma could be used as a research tool for preclinical screening of new drugs.
