Background and Objective:6-phosphofructo-2-kinase 3(PFKFB3)is a key rate-limiting enzyme in cell glycolysis pathway.Previous studies have demonstrated that PFKFB3 is closely related to the occurrence and development of many malignant tumours and regulates the biological behaviors of tumour cells.This study aims to investigate the expression level of PFKFB3 in gastric cancer tissues and its role in the proliferation and apoptosis of gastric cancer cell and the internal molecular biological mechanism.Methods:1.The TCGA database was used to analyze the expression level of PFKFB3 in gastric cancer tissues and adjacent normal tissues,and the relationship between the PFKFB3 expression and the prognosis of gastric cancer patients.2.Quantitative real-time PCR,Western blotting,and immunohistochemistry assays were used to analyze the expression level of PFKFB3 in gastric cancer tissue samples and the adjacent normal tissue samples.3.After down-regulating the PFKFB3 expression in gastric cancer cells,CCK8 and EdU assays were used to observe cell proliferation ability.Flow cytometry was used to detect cell apoptosis rate and cell cycle changes.4.The effect of PFKFB3 on PI3 K / AKT signaling pathway was analysed by Western blotting assay.After overexpressing PFKFB3 expression and using PI3K/AKT pathway inhibitor,gastric cancer cell proliferation was detected by CCK8 assay.Results:1.TCGA database analysis showed that the expression level of PFKFB3 in gastric cancer tissues was significantly higher than that in adjacent normal tissues(P <0.05),and the high expression of PFKFB3 was closely related to the poor prognosis of patients with gastric cancer.2.Real-time quantitative PCR,Western blotting and immunohistochemistry assays further confirmed that PFKFB3 expression was significantly increased in gastric cancer tissues compared with adjacent normal tissues(P <0.01).PFKFB3 expression was closely associated with lymph node metastasis and clinical TNM stage(P <0.05).3.After PFKFB3 downregulation in gastric cancer cells,cell proliferation was significantly decreased(P <0.01),while cell apoptosis significantly increased(P <0.01)and the G1-S phase progression of cell cycle was blocked(P <0.01).4.Down-regulated the expression level of PFKFB3 in gastric cancer cells.Total PI3 K and AKT did not change,while phosphorylated PI3 K / AKT decreased.After overexpressing PFKFB3 expression and using PI3K/AKT pathway inhibitor,gastric cancer cell proliferation was significantly inhibited.Conclusion:1.The expression level of PFKFB3 in gastric cancer tissues is significantly increased compared with corresponding adjacent normal tissues,and patients with high PFKFB3 expression had poor prognosis.2.Down-regulating the PFKFB3 expression can significantly inhibit cell proliferation and cell cycle progression,while increase the apoptotic rate of gastric cancer cells.3.PFKFB3 regulates the growth of gastric cancer by affecting phosphorylated PI3 K / AKT signaling pathway.
