Objective:To explore the relationship between YAP/TAZ protein and the occurrence and development of colon cancer under the hippo signal pathway,and to make statistical analysis with the general clinical data of patients,so as to provide new ideas for the search for new treatment methods of colon cancer.Methods:From January 2017 to December 2019,pathological tissue samples of 30 patients with colorectal cancer undergoing radical operation were collected from 908Hospital of the joint service support force,including cancer tissue and adjacent tissue(more than 2cm from the edge of cancer tissue,he)The expression of Yap/TAZ,CD44/CD24 and MMP9/MMP2 in pathological samples were detected by immunohistochemistry.The expression data of various proteins in colon cancer and adjacent tissues were collected and analyzed statistically.Results:The positive rates of YAP/TAZ,CD44/CD24 and MMP9/MMP2 were 83.33%,63.33%,53.33%,66.67%,53.33%and 46.67%respectively in cancer tissues and 13.33%,10%,6.67%,16.67%,20%and 13.33%in paracancer tissues.There was a significant positive correlation between YAP expression and TAZ expression in cancer tissues(?~2=10.4,P=0.001<0.05,R=0.588,P=0.001<0.05);there was a significant positive correlation between YAP expression and CD44 expression(?~2=6.86,P=0.009<0.05,R=0.478,P=0.008<0.05);there was a significant positive correlation between YAP expression and CD24 expression(?~2=12,P=0.001<0.05,R=0.632,P=0.001<0.05).There was a significant positive correlation between the expression of TAZ and YAP(?~2=10.4,P=0.001<0.05,R=0.588,P=0.001<0.05);there was a significant positive correlation with the expression of CD44 protein(?~2=13.7,P=0.001<0.05,R=0.675,P=0.001<0.05);there was a significant positive correlation with the expression of CD24 protein(?~2=7.18,P=0.007<0.05,R=0.489,P=0.006<0.05);there was a significant positive correlation with the expression of MMP9 protein(?~2=13.7,P=0.001<0.05,R=0.675,P=0.001<0.05).In 30 cases of colon cancer,there were 20 cases of poorly differentiated adenocarcinoma,of which 19 cases were positive for YAP/TAZ protein,and 1 case was negative for YAP protein.In 10 cases of differentiated adenocarcinoma,5 cases were negative for YAP/TAZ protein expression at the same time,and 5 cases were negative for Yap protein expression.It can be seen from table 8 that the expression of YAP protein is positively correlated with the pathological classification of colon cancer(?~2=12.0,r=0.632,P=0.001<0.05);the expression of TAZ protein is positively correlated with the pathological classification of colon cancer(?~2=25.9,r=0.929,P=0.001<0.05).In 30 cases of colon cancer,21 cases had a maximum tumor diameter of more than 4cm,of which 19 cases were positive for YAP/TAZ protein,and 2 cases were negative for YAP protein.In the remaining 9 cases of colorectal cancer with the largest diameter less than 4cm,5 cases were negative for YAP/TAZ protein expression at the same time,and 4 cases were negative for YAP protein expression.The results showed that the expression of YAP was positively correlated with the tumor size(?~2=14.0,r=0.683,P=0.001<0.05);the expression of TAZ was positively correlated with the tumor size(?~2=22.2,r=0.860,P=0.001<0.05).Among the 30 pathological specimens of colon cancer,20 cases had mesenteric lymph node metastasis,19 cases were positive for YAP/TAZ protein,and 1 case was negative for YAP protein.Among the 10 pathological specimens without metastasis of mesenteric lymph node,5 were positive for YAP protein and 5 were negative for YAP protein.The results showed that the expression of YAP was positively correlated with the metastasis of colorectal cancer(?~2=12.0,r=0.632,P=0.001<0.05);the expression of TAZ was positively correlated with the metastasis of colorectal cancer(?~2=25.9,r=0.929,P=0.001<0.05).Conclusion:The expression of YAP/TAZ,CD44/CD24 and MMP9/MMP2 in colon cancer was significantly higher than that in adjacent tissues.Combined with the general clinical data of patients,it was found that hippo YAP/TAZ protein plays an important role in the development of colon cancer.Its mechanism may be through the regulation of CD44/CD24 protein in cancer stem cells and MMP9/MMP2 protein in matrix metalloproteinases.
