Objective: To observe the changes in the gut microbiota of patients with primary biliary cholangitis(formerly known as primary biliary cirrhosis)(Primary Biliary Cholangitis,PBC),and whether the gut microbiota changed after ursodeoxycholic acid treatment.Explore and analyze the mechanism of gut microbiota in the pathogenesis of PBC to provide clinical data.Methods: Using the latest PBC guidelines as the standard for diagnosis and treatment of PBC,the patients who were diagnosed as PBC in the First Affiliated Hospital of Shihezi University from November 2018 to December 2019 were collected,and a simple random sampling method was used to select 5 PBC The patient collected blood biochemical indexes and feces before and 3 months after the treatment of Ursodeoxycholic Acid(UDCA)and collected blood biochemical indexes,5 biomarkers of the same period in the same physical examination group with similar gender,age and BMI Feces,genomic DNA is extracted,and metabolic sequencing technology is used for gut microbiota analysis.Results: After 3 months of UDCA treatment,clinical indicators such as AST,ALT,ALP,and GGT of PBC patients were relieved,and the difference was statistically significant(P<0.05);there was no change in gut microbiota diversity in PBC patients and healthy groups Significant difference(P>0.05).At the phylum level,PBC patients and healthy groups had the highest abundance of Firmicutes Firmicutes,Bacteroidetes,Proteobacteria,and Actinobacteria in the phylum.Compared with the healthy group,PBC patients were enriched in the genera of Arsenophonus,Jerseyvirus,Tetzosporium,Alkalibacillus,Tatlockia,Phormidesmis,Aerobacillus,Waddlia,Gillisia,etc.,and the difference was statistically significant(P <0.05);at the species level,Compared with PBC patients and healthy groups,they were enriched in Bacteroides_sp,Clostridium_sp,Clostridium_sp,Roseburia_sp,Lactobacillus_sp,Bacillus_sp,and Prevotella_sp(P<0.05);after UDCA treatment,compared with before treatment,PBC patients in Clostridium_sp,Enterococcus_sp,Amphritea_sp,Bacillus_sp,Liszt Listeria_seeligeri,Aeromonas_sp and Pantoea_allii were enriched(P<0.05);while the abundance of bacteria such as Pseudomicrostroma_glucosiphilum,Neisseria_weaveri,Dissulfuribacter_thermophilus and other bacteria decreased(P<0.05),and some bacterial groups were reversed before treatment.Conclusion: Compared with healthy people,PBC patients have changed gut microbiota.After UDCA treatment,not only liver function has been improved,but some gut microbiota imbalance have been reversed.Gut microbiota may be a potential target for PBC treatment.
