Objective:To investigate the influence and mechanism of erythropoietin on pulp revascularization in immediate reimplanted immature molars rats,and to elaborate the expression of vascular endothelial cell growth factor on pulp healing in immediate reimplanted immature molars rats.Methods:Eighty 4-week-old male healthy Wistar rats with specific pathogen free(SPF)were arbitrarily divided into four groups according to different drugs: the blank control group(non-tooth extraction),the negative control group(normal saline),the positive control group(gentamicin)and the observation drug group(EPO).Each group has twenty rats.Before replantation,each group was dealt with its relevant solution for 4 minutes.The rats were killed on days 3,7,14,21 and 28 individually,then the specimens were made in the surgical area.Hematoxylin-eosin staining was used to explore the pulp reconstruction in different time periods.Immunohistochemical staining was used to study the expression levels of VEGF in pulp tissue of the tooth at different times in each group rats.Results:The hematoxylin-eosin staining results reveal that comparing withnon-tooth extraction group,inflammation in the pulp tissue of replanted teeth in normal saline group is heavier,the development of the root is bad,the restorative dentin and cementum is less,and bigger apical pores;the inflammation of the rats in gentamicin and EPO group was mild,at the same time,the development of the root was relatively better;there were more restorative dentin and cementum,and the more narrowed apical pores.The immunohistochemical results reveal that,comparing with non-tooth extraction group,the VEGF immunoreactivity of replanted teeth demonstrated strong positive staining on the 3rd,7th and 14 th day in the other groups.Subsequently,the staining gradually weakened with the time passed.The average optical density(AOD)of VEGF in positive area demonstrated that EPO group > gentamycin group > normal saline group > non-tooth extraction group.Compared with Unoperated group,there was significant difference in normal saline group,gentamicin group and EPO group at 3,7,14 and 21 after the surgery(P < 0.05),while there was no significant difference of VEGF in tissue of the replanted teeth in rats at 28(P > 0.05).Comparing with normal saline group,the expression levels of VEGF in pulp tissue of the replanted teeth in gentamicin group and EPO group at 3,7,14 and 21 after surgery had significant differences(P < 0.05),but the expression of VEGF in tissue of the replanted teeth in rats at 28 have no significant differences(P > 0.05).Comparing with gentamicin group,at every time points,there were no significantdifferences in pulp tissue of the replanted teeth in rats in EPO group(P>0.05).Conclusion:EPO could raise the expression of VEGF,increase the angiogenesis in pulp tissue of the replanted teeth,and furnish rich vascular bed for replantation,as a result,exerting the potential of dental pulp repair and defense,and promote the healing of the replantation.
