ObjectiveIn this study,high-throughput proteomics was used to screen out the biological protein factors that changed in serum of OSAHS patients compared with those of healthy people. Then, the sample size was expanded to evaluate the relationship between the expression of these protein factors in serum and the severity of OSAHS,and the relationship between these protein factors and PSG parameters was analyzed To explore the pathogenesis of OSAHS and its relationship with hypertension and lipid metabolism in order to provide guidance for clinical treatmentMethod:Part ?:Quantitative analysis of proteomics in serum of OSAHS patients and control group:From August 2018 to December 2018,10 cases of OSAHS adult male patients were selected from the Department of Otolaryngology, the second hospital of Jilin University. At the same time,10 cases of healthy control group adult male patients were selected. The ratio of up-regulation and down-regulation of serum bioproteins in OSAHS patients was calculated. The up-down factors related to the pathogenesis of OSAHS were foundPart?:Correlation analysis of serum SOD,CAT expression and OSAHS:from December 2018 to November 2019,33 patients with OSAHS were selected from the Department of Otolaryngology, head and neck surgery, the second hospital of Jilin University.According to the results of PSG,33 patients with moderate and severe degree were selected, and 25 healthy control group were selected from the physical examination center of our hospital. The concentration of SOD and CAT in the serum was detected by colorimetry. The difference was statistically significant with SPSS 20.0(P<0.05)Part ?:Dyslipidemia and OSAHS:from December 2018 to November 2019,33 cases of OSAHS patients were selected from the otorhinolaryngology head and neck surgery department of the second hospital of Jilin University.25 cases of the control group were selected in the physical examination center. The diagnosis standard of dyslipidemia refers to the standard of dyslipidemia formulated by the Chinese adult dyslipidemia prevention and control committee.Blood lipid was detected by the laboratory of the second hospital of Jilin University. The statistical method is the same as the second part. The detection of serum SOD and CAT is the same as the second part.Part ?:Hypertension and OSAHS:33 cases of OSAHS patients were selected from the otorhinolaryngology head and neck surgery department of the second hospital of Jilin University from December 2018 to November 2019.25 cases in the control group were selected from the physical examination center. The diagnosis standard of hypertension was referred to the Chinese hypertension diagnosis Guide(2018 Revision). The statistical method is the same as the second part. The detection of serum SOD and CAT is the same as the second partResult:Part ?:Protein differential expression analysis showed that there were 32 up-regulated proteins and 44 down-regulated proteins. The downregulated protein SOD and CAT were selected for further sample size analysisPart ?1.Basic data analysis between the OSAHS group and the control group:the age distribution of the two groups was approximately the same(40.55 ± 10.60/40.92 ± 9.04,P=0.89),and there was no significant difference between the two groups.BMI(30.0015.56/26.4013.04,P=0.005),P<0.05,the difference was statistically significant2.The expression of SOD and CAT in OSAHS group and control group:SOD(90.81 ±17.12/98.60±6.64,P=0.00),CAT(14.50±13.42/37.61 ±25.33,P=0.01),P<0.05,the difference was statistically significant.3.Correlation analysis of serum SOD and CAT levels with age,BMI and PSG detection results:there was a negative correlation between serum SOD and AHI(r=-0.43,P=0.01).Part ?:1.The general data, blood lipid and metabolic indexes of OSAHS group and control group were compared and analyzed:The analysis of ages and BMI are the same as part ?,the concentrations of uric acid,TG,TC,HDL-C,SOD and CAT in serum were(3.46±0.86/2.99± 0.84,P=0.05),(2.70± 1.52/1.86± 1.29,P=0.04),(5.32± 0.92/4.64± 0.82,P=0.00),HDL-C(1.02± 0.19/1.14± 0.20,P=0.03).LDL-C(3.46± 0.86/2.99± 0.84,P=0.05)?SOD(90.81 ±17.12/98.60± 6.64,P=0.02)?CAT(14.50 ± 13.42/37.61 ± 25.33,P=0.00),with statistical significance;as for GGT(67.47± 46.24/47.57 ± 36.88,P=0.08),there was no significant statistical difference.2.Two Analysis of risk factors in OSAHS normal group and OSAHS abnormal group:the concentrations of GGT,uric acid and TG in serum were(48.18 ±5.07/77.36 ± 11.13,P=0.02),(1.85 ± 0.13/2.93 ± 0.33,P=0.00),(4.69 ± 0.10/5.46±0.20,P=0.00),with statistical significance;age(P=0.95),BMI(P=0.94),TC(P=0.59),HDL-C(P=0.65),LDL-C(P=0.59),SOD(P=0.07),CAT(P=0.30),AHI(P=0.42),LaS02(P=0.12),CT90(P=0.58),there was no significant statistical difference.3.Correlation analysis of metabolic related indexes with age,BMI and PSG monitoring results:TC was positively correlated with SOD(r=0.42,P=0.02),LDL-C was positively correlated with CAT(r=0.42,P=0.02),and no significant correlation was found.Part ?:1.Comparison of the number of hypertension cases between OSAHS group and control group:the rate of hypertension in OSAHS group was 64%(24/33),40%(10/25),x=6.28,P<0.05,with statistical significance2.Analysis of hypertension group and non-hypertension group in OSAHS group and control group:comparison of OSAHS with hypertension group and OSAHS without hypertension group:age(43.33± 10.85/35.67 ± 8.50,P=0.04),SOD(85.04 ± 8.49/95.98± 14.04,P=0.01),CT90(21.34 ± 18.10/42.02 ± 27.87,P=0.02),P<0.05,with statistical significance;no significant difference in other results.Conclusion:1.Compared with the normal control group, there are protein expression differences in the serum of OSAHS patients, which may be related to the pathogenesis of OSAHS2.There is imbalance of antioxidation in OSAHS patients, which leads to oxidative stress.Oxidative stress may be related to the degree of hypoventilation and hypoxia in OSAHS patients3.There is metabolic disorder in OSAHS patients. The oxidative stress response induced by IH may be related to the metabolism of blood lipids.Excluding age, weight and other factors,OSAHS itself may cause metabolic abnormalities4.Oxidative stress in OSAHS may be related to hypertension, which is a factor of hypertension in OSAHS.
