Objective:The aim of the study was to compare the different expression of HMGA2 protein among papillary thyroid carcinoma,thyroid follicular adenoma and normal thyroid tissue,so as to clarify its value in differential diagnosis of benign and malignant thyroid nodules and further explore the relationship between the expression of HMGA2 protein and the metastasis of cervical lymph nodes in papillary thyroid carcinoma.Methods:125 thyroid samples including 85 PTC,20 FTA and 20normal thyroid tissue were retrospective analyzed.Expression of HMGA2protein was compared among these three groups.Furthermore,PTC group was subdivided according to the TMN staging and the number of metastasis lymph nodes in order to explore the feasibility of HMGA2 protein in predicting the lymph node invasion of PTC.In addition,the efficacy of BRAFV600E and HMGA2 in the determination of cervical lymph node metastasis was compared.RESULTS:The rate of the expression of HMGA2 protein in PTC was100%,which was statistically different from that of thyroid adenoma and normal tissue?P<0.05?.Moreover,there was no significant correlation between expression of HMGA2 protein and TNM staging,the size of PTC or whether it was accompanied by Hashimoto's thyroiditis.But it was associated with T staging,N staging and the number of metastasis lymph nodes.Furthermore,HMGA2 is more sensitive in the prediagnosis of cervical lymph node metastasis than BRAFV600E.Conclusions:?1?HMGA2 protein can be used as a molecular marker for differential diagnosis of benign and malignant thyroid nodules.?2?The expression level of HMGA2 protein is positively correlated with cervical lymph node metastasis in papillary thyroid carcinoma,which shows certain auxiliary significance for clinical decision.
