OBJECTIVETo establish TRALI rat models induced by homologous plasma and allogeneic plasma respectively,and compare the clinical manifestations and pathological changes of lung tissue with those of TRALI models successfully established by human plasma induction,and explore the feasibility and validity of models induced by homologous plasma and allogeneic plasma respectively.METHODSMale Sprague Dawley rats(SD rat)are used to establish animal models.SD plasma group was given intraperitoneal injection of 2 mg/kg LPS,2 hours later,1 ml of blood was removed from femoral vein and 1 ml of SD rat plasma was transfused;male Brown Norway rat(BN rat)plasma group was given intraperitoneal injection of 2 mg/kg LPS,2 hours later,1 ml of blood was removed from femoral vein and 1 ml of BN rat plasma was transfused;human plasma group was given intraperitoneal injection of 2 mg/kg LPS,2 hours later,1 ml of blood was removed from femoral vein and 1 ml of human plasma was transfused.Control group was given intraperitoneal injection of 2 mg/kg LPS,after 2 hours,1 ml of blood was removed from femoral vein and 1 ml of normal saline was infused.Positive group was given intraperitoneal injection of 2 mg/kg LPS,and 2 hours later,1 ml of blood was removed from femoral vein and 5 ml/kg of LPS was infused.After the experiment,the right middle lobe of the rat lung was taken to calculate the dry-wet weight ratio of the lung tissue.At the same time,the lung tissue was taken for H&E staining.According to the changes of the lung tissue structure,the pathological score of lung injury was performed.The level of brain natriuretic peptide in plasma was measured before and after the experiment.The wet-dry weight ratio of lung tissue,pathological score of lung injury and the level of brain natriuretic peptide were analyzed statistically.RESULTSLung injury occurred in SD rats and BN rats in pathology,but the clinical symptoms of SD plasma group were not obvious.The wet-dry ratio of lung tissue between SD and BN plasma group were significantly different from those in human plasma group(t=0.525,p=0.002;t=0.297,p=0.048),so were the pathological scores(t=4.8,p=0.000;t=2.2,p=0.046).There was significant difference in pathological scores of lung injury between BN plasma group and SD plasma group(t=2.6,p=0.022).CONCLUSIONIn this study,SD rats with LPS 2 mg/kg intraperitoneally injected and 10% total blood volume of BN plasma infused were more effective than SD plasma in inducing TRALI.There are still differences between the TRALI model induced by BN and human plasma.Therefore,the results of this study confirm that human plasma infusion is still the best way to establish rat TRALI model.
