Terahertz time-domain spectroscopy and infrared micrspectroscopy technology are playing an important role in biomedical research.It is a significant way to industrialization of spectroscopy technology to exploring the spectral characteristics of different biological tissues or cells.For the eye is the key part to feel the outside information,the tissue and cells of rat eyeball were used as the study object.Considering high brightness and high resolution of the synchrotron radiation light source,the synchrotron radiation infrared microscopic imaging were used.The results imply that terahertz near-filed imaging and infrared microscope spectroscopy technology is important to understand the mechanism and the early diagnosis of disease.The main work of this study is as follows:1.The terahertz near-field imaging and synchrotron radiation infrared microscopic imaging were performed on the paraffin embedded and frozen eyes of mice,and the terahertz and infrared spectra of mouse eyeballs were obtained.The terahertz and infrared absorption spectra of the samples were analyzed based on the structure and composition of the eyeball.2.Synchrotron radiation infrared microimaging was used to study the freeze-dried retina of mice.The infrared spectra of mouse retina and the distribution of different molecular vibrational or rotation modes in the retina were obtained.And the infrared spectrum and second derivative spectrum of retina were analyzed.The results show that terahertz spectrum and infrared spectrum can be used in fundus examination.3.TGF-? was used to induce the excessive proliferation of trabecular cells in vitro to simulate one of the main causes of primary open-angle glaucoma.Then the properties of terahertz spectrum and infrared spectrum of cells before and after induction were checked,and the spectral characteristics of the cells were analyzed in terms of the changes of the main substances.The results provide a basis for the detection of THz and IR spectra of primary open angle glaucoma and other diseases.
