Objective:To investigate the effects of sevoflurane on the Parthanatos of HT22 mouse hippocampal neuron cells and hippocampal neurons in neonatal rats,and to clarify the possible mechanism of developing cerebal neurons death induced by sevoflurane.Methods:HT22 mouse hippocampal neuron cells were treated with 2%,4% and 8% sevoflurane for 6,12 and 24 h.HT22 mouse hippocampal neuron cells were pretreated with 3-Aminobenzamide(3AB)or normal saline for 1 h and treated with 4% and 8% sevoflurane for 12 h;seven-day-old neonatal rats were treated with 2.5% sevoflurane for 6 h.HT22 mouse hippocampal neuron cells survival and mortality were detected by MTT and LDH methods,the contents of PARP-1 signaling pathway related proteins in HT22 mouse hippocampal neuron cells and hippocampus of neonatal rats were detected by Western blot,changes of mitochondrial membrane protein were detected by jc-1 fluorescence staining.Results:Sevoflurane induces HT22 mouse hippocampal neuron cells death in a dose-time dependent manner(P<0.01),reduces cell survival(P<0.01),decreases mitochondrial membrane protein,promotes the expression of PARP-1,PAR and AIF proteins in HT22 mouse hippocampal neuron cells and hippocampus of neonatal rats(P<0.01).3AB significantly reduces HT22 mouse hippocampal neuron cells death induced by sevoflurane(P<0.01),promotes cells survival(P<0.01),inhibitings the decrease of mitochondrial membrane protein induced by sevoflurane,decreases the expression of PARP-1,PAR and AIF in HT22 mouse hippocampal neuron cells(P<0.01).Conclusion:Sevoflurane Induces Parthanatos of HT22 mouse hippocampal neuron cells and hippocampal neuron in neonatal rats through activating PARP-1-dependent death signaling pathways.
