Research objectives:To explore the expression levels of LncRNA H19 and IGF2 in breast cancer,the role of LncRNA H19 and IGF2 imprinting loss in breast cancer and the correlation between LncRNA H19 and IGF2 in breast cancer pathology.Research methods:1.The expression of lncRNA H19 and IGF2 in breast cancer and its adjacent tissues were detected by RT-PCR.2.IGF2(Apa I site)and H19(Alu I site)fragments were amplified by PCR,and the gel was recovered and purified for sequencing.SNP was used to distinguish expression of alleles.The imprinting status of lncRNA H19 and IGF2 in breast cancer was detected by gene sequencing,and the loss of imprinting effect of LncRNA H19 and IGF2 expression was analyzed.3.To analyze the relationship among the loss of imprinting of lncRNA H19 / IGF2 in breast cancer tissue,the clinical classification and pathological information.Meanwhile,the relationship between lncRNA H19 and IGF2 in breast cancer was investigated.Results:1.The expression level of lncRNA H19 and IGF2 in breast cancer(120 cases and 134 cases)was higher than that in adjacent tissues by RT-PCR.Correlation analysis showed that there was a positive correlation between the expression level of LncRNA H19 and IGF2 in breast cancer,r = 0.567(P = 0.00011).The expression level of LncRNA H19 in breast cancer was(0.088±0.014),which was significantly higher than that of LncRNA H19 in breast cancer tissues(0.024 ± 0.005);the expression level of IGF2 in breast cancer tissues was(0.348 ± 0.036),which was significantly higher than that of IGF2 in breast cancer tissues(0.095 ± 0.0183).2.Both lncRNA H19 and IGF2 have loss of imprinting in breast cancer.The frequency of IGF2 LOI was 36.7%(22 cases of LOI and 38 cases of MOI,22/60),which was significantly higher than that of LncRNA H19(2 cases of LOI and 45 cases of MOI,2/47).3.The expression of IGF2 in the group of loss of imprinting was significantly higher than that in the group of maintenance of imprinting.The expression level of LOI-C(22 cases)was(0.127±0.226)and MOI-C(38 cases)was(0.053 ± 0.156)in breast cancer.There were few cases(2 cases)of LncRNA H19 loss of imprinting.Conclusion:1.The expression level of LncRNA H19 and IGF2 in breast cancer is higher than that in the adjacent tissues,which is positively correlated with each other,providing a new molecular marker for the diagnosis of breast cancer.2.There were LncRNA H19 and IGF2 loss of imprinting in breast cancer.The incidence of IGF2 imprint loss was significantly higher than that of LncRNA H19.3.The expression level of IGF2 loss of imprinting is significantly higher than that maintenance of imprinting,so LOI is one of the important factors leading to breast cancer,which provides a new direction for gene therapy of breast cancer.
