Objective: To explore and analyze the different roles of epithelial mesenchyma transformation(EMT)in the pathogenesis of non-small cell lung cancer(NSCLC)and chronic obstructive pulmonary disease(COPD),and thus to provide a new theoretical basis for the pathogenesis,prevention and targeted therapy of these two diseases.Methods: 1.According to the inclusion criteria and exclusion criteria,alveolar lavage fluid samples were collected from patients diagnosed with NSCLC,COPD,and COPD complicated with NSCLC.The above samples were centrifuged and the RNA of the precipitated cells was extracted with Trizol reagent.2.Four samples were taken from each of the three groups,and the differential genes were screened out by PCR Array.Then,q RT-PCR was used to verify the expression of the differential genes ITGB1,VIM,KRT14,mmp-9 and Notch1 in the three groups of samples.3.All data of these differential gene expressions in the three groups of disease samples were expressed as mean ± standard deviation(sx ±),and statistical analysis was performed using SPSS19.0 software.Results: ITGB1,VIM and MMP-9 were significantly up-regulated in the NSCLC group(P<0.05).ITGB1,VIM,MMP-9 and Notch1 were up-regulated(P<0.05),while KRT14 was down-regulated(P<0.05)in patients with COPD complicated with NSCLC.In the NSCLC sub-group,MMP-9 and Notch1 were highly expressed in the lung adenocarcinoma group compared with in the squamous cell lung carcinoma group(P<0.05).Conclusion: 1.The key genes involved in EMT in non-small cell lung cancer are ITGB1,VIM,MMP-9,Notch1 and KRT14.2.Up-regulation of ITGB1,VIM,MMP-9,Notch1 and down-regulation of KRT14 may be more closely related to the occurrence and development of NSCLC in patients with COPD complicated with n NSCLC.3.In the NSCLC sub-group,MMP-9 and Notch1 genes were highly expressed in the lung adenocarcinoma group compared with the lung squamous cell lung carcinoma group,and these two genes may be more closely related to the occurrence and development of lung adenocarcinoma.
