Objective: To explore the establishment of the model which accords with the observation of the course of the Excess Syndrome of Yangming Fu-Organs and the study of the changes of the inflammatory factors related to Yang Ming disease(Excess Syndrome of Yangming Fu-Organs).Methods:(1)The experimental rats were randomly divided into control group,high dose model group,medium dose model group and low dose model group.Different doses of LPS were injected.Body temperature,mortality rate,leukocyte count by whole blood cell counter,plasma LPS and small intestine IL-6 were measured by enzyme-linked immunosorbent assay.(2)The experimental rats were randomly divided into the solved control group,the solved model group,the control group and the model group.After establishing the Yang Ming diease model,blood and intestinal tissues were taken every day for 5 consecutive days,blood routine was detected by blood cell analyzer,TLR4 Protein content was measured by immunohistochemistry,real-time quantitative aggregation by fluorescence The level of TLR4 m RNA was detected by PCR.Results:(1)the body temperature,plasma LPS and intestinal mucosa IL-6 content of each model group were higher than those of the normal group.On the 5th day after the model was established successfully,the leukocytes and neutrophils in the dose group increased significantly,and the mortality of each dose model group was in direct proportion to the LPS injection amount.(2)The number of leukocytes,neutrophils and monocytes in the solved model group was higher than that in the model group,the number of monocytes increased significantly(P < 0.05),TLR4 m RNA expression and protein content in the solved model group were significantly lower than that in the non model group on the first day(P < 0.05).Conclusion: 5 mg / kg LPS injection dose is the effective injection dose to observe the course change model of the Excess Syndrome of Yangming Fu-Organs.In the course of f the Excess Syndrome of Yangming Fu-Organ,the difference of neutrophils and lymphocytes in the solved model group was earlier and longer than that in the control group,which means that the immune response was more obvious in the solved model group.TLR4,as an important receptor upstream of the inflammatory pathway,was stimulated by LPS,and TLR4 m RNA and protein expression were significantly up-regulated,but the level of TLR4 m RNA and protein in the model group was lower than that in the solved model group,indicating that the inflammatory response of Excess Syndrome of Yang-Ming Fu-Organs in the model group was lighter than that in the model group,which may be related to the decreased release of inflammatory mediators.
