Objective: Explore the relationship between pathological changes in peripheral tissues and the infiltration,growth and metastasis of HAE,Pathological features of peripheral tissues in different stages and different distance of hepatic echinococcosis were studied by HE stain and immunohistochemistry.Methods: Building the model of rat HAE diseases.Tissues were taken at different distances from the lesion after 12,24,36 and 48 weeks respectively.HE stain to observe the cell morphology.The expressions of VEGF-R2?CD34??-SMA??-cat?VIM?OPN?MMP-2 in tissues were determined by immunohistochemistry,and the statistical description and analysis were performed.Results:(1)At the 12 th week after modeling,there were about 3 mm alveolar lesions at the puncture point of liver modeling,which were pale white.After that,the lesions gradually enlarged,and at week 24,36 and 48,multiple cystic solid masses were present.(2)HE staining showed numerous inflammatory cell infiltrations in tissues 0.5 cm away from the lesion.With the extension of time after modeling,the infiltration range of inflammatory cells increased.HE stain at a distance of 1.0 cm from the lesion and beyond showed no obvious abnormalities.No micro vesicles were seen in all tissues.(3)The expression of VEGF-R2 gradually increased and the positive expression range also increased.At week 24,the positive expression in tissues 0.5 cm away from the lesion increased.The positive expression limit in 1.0 cm and 1.5 cm away at week 36 th and 48 th respectively.The expression of CD34 was mainly located in vascular endothelial cells and hepatic sinus.At the 36 th and 48 th week,the tissue expression was increased within the range of 0.5 cm from the lesion.The ?-SMA was mainly expressed in the hepatic sinus and the portal area.Positive expression was observed in the cyst wall of the lesion.There was no significant difference between the expression of distance and normal liver in each period.The ?-cat was mainly localized in cytoplasm and cell membrane,and no significantly expressed in the tissues at the distance from the lesion at week 12 and 24.At the 36 th and 48 th week,it was mainly expressed in the range of 1.0 cm from the lesion.VIM was positively expressed to different degrees in tissues 0.5 cm away from the lesion at each stage,with significant differences among individuals.The express of OPN in the 12 th week,there were no significant increase in positive expression was observed in all the regional tissues.In the 24 th week,positive expression was found in tissues 0.5 cm away from the lesion.In the 36 th and 48 th week,positive expression was found in tissues 1.0 cm away from the lesion.No significant increase in positive expression of MMP-2 was observed in all regional tissues at week 12 and 24.At the 36 th week and within the range of 1.0 cm from the lesion,the tissue positive expression was increased.Conclusion:(1)After modeling,the surrounding tissues of rat HAE showed angiogenesis,EMT and changes in the extracellular matrix.(2)Changes in angiogenesis,EMT and extracellular matrix mainly occurred in the middle and late stages of the disease,and there were significant individual differences in EMT.
