Objective: To investigate the distribution characteristics of serum folic acid,homocysteine level and MTHFR C677 T gene polymorphism in preeclampsia patients in xining,qinghai province,and to provide theoretical and clinical basis for early diagnosis and timely prevention of preeclampsia patients.Method: Using stratified randomized grouping case research method,collecting 100 cases between September 2017 and October 2019 in qinghai province people’s hospital for prenatal diagnosis of preeclampsia women as observation group(preeclampsia group),at the same time to collect 100 cases with the same period(the same as the observation group of pregnant days)in qinghai province people’s hospital of the health of pregnant women as control group(normal pregnancy group).General information of all subjects included in the clinical trial was collected and recorded: age,pregnancy,delivery,pregnancy days,family history of hypertension,weight,sitting blood pressure,urine protein.Peripheral venous blood of all the selected subjects was collected and serum folic acid(FA)levels were detected by high performance liquid chromatography(HPLE).Serum homocysteine(Hcy)levels were detected by enzyme linked immunosorbent assay(ELLSA).Polymerase chain reaction(PCR)was used to detect the gene polymorphism at the MTHFR C677 T site.General data of control group and observation group were recorded.The distribution characteristics of serum folic acid,homocysteine levels and MTHFR C677 T gene polymorphism were also recorded.The recorded data were processed by SPASS20.0 statistical software and analyzed in parallel.Serum concentrations of folic acid and homocysteine in healthy pregnant women and patients with preeclampsia in xining,qinghai were compared,and the genotype distribution characteristics of MTHFR in the blood of the two groups were compared.Results:1.General comparison of the two groups of objects.There was no difference in age between the control group and the observation group(P>0.05);There was no difference in pregnancy and delivery between the control group and the observation group(P>0.05);There was no difference in pregnancy days between the control group and the observation group(P>0.05);There was no difference in family history of hypertension between the control group and the observation group(P>0.05);There was a difference in body weight between the control group and the observation group,and the weight of the observation group was higher than that of the control group(P<0.05);There were differences in sitting blood pressure between the control group and the observation group(P<0.05),the average blood pressure of the observation group was higher than that of the control group;There were differences in urinary protein between the control group and the observation group(P<0.05),urine protein was negative in the control group and positive in the observation group;2.Comparison of serum folic acid levels between the two groups.Comparison of serum folic acid levels between the control group and the observation group showed that serum folic acid levels in the control group were normal,and serum folic acid levels in the observation group were reduced on average(P<0.05);3.Comparison of serum homocysteine levels between the two groups.Comparison of serum homocysteine levels between control group and observation group showed no difference(P>0.05);4.Comparison of serum MTHFR C677 T gene polymorphism distribution between the two groups.The comparison of the polymorphism distribution of serum MTHFR C677 T locus in the control group and observation group showed that the expression of serum MTHFR C677 T locus in the control group was mostly CC type,while the expression of serum MTHFR C677 T locus in the observation group was mostly CT type and TT type.Conclusion:1.Weight gain,increased sitting blood pressure,and increased urinary protein in patients with preeclampsia in xining area are consistent with the clinical manifestations and basic diagnostic criteria of patients with preeclampsia;2.The serum folate level of preeclampsia patients in xining area was reduced,which provided the treatment idea and basis for such patients from the perspective of folic acid supplementation;3.The homocysteine level of preeclampsia patients in xining region is not high,so whether the homocysteine level of preeclampsia patients in xining region is specific or not remains to be further studied;4.The gene polymorphism at the MTHFR C677 T locus in the serum of preeclampsia patients in xining was distributed in CT type and TT type,which is of great significance for the prevention,treatment and diagnosis of preeclampsia in advance.
