Abstract/Summary:
Objectives:The model was established by implanting 5%-HA / PLA composite root canal post into beagle dogs’ deciduous canine and deciduous molar.The morphology and histological structure of the inherited permanent teeth were studied by clinical examination,histological HE staining,tooth grinding,pulp stem cell culture and osteogenic and lipogenic induction methods,Whether the morphology of dental pulp cells and the differentiation of dental pulp stem cells have an effect,so it can be concluded that PLA / HA complex has no effect on the inherited permanent teeth.Methods:1.(1)experimental group: two 3-month-old beagles were selected for pulpotomy.After pulpotomy,the antibiotic paste in the upper part of the root canal was removed and kept for at least 4 mm.The modified PLA /ha root canal post was implanted into the experimental group with glass ionomer filling.(2)Control group: the right homonymous teeth were not treated,and the animal model was established for the control group.2.After the animal model was established,the experimental animals were divided into two groups.(1)Vital signs,teeth,gums and looseness were examined after operation and before extraction.(2)The first group of beagle dogs were perfused and fixed by common carotid artery 4 weeks after operation.The experimental group and the control group were taken and examined by HE staining.(3)In the second group,8 weeks after the operation,the experimental group and the control group were selected to inherit the permanent canine and the second molar respectively.The canine was dehydrated by gradient,and the hard tissue abrasive film was made after embedding(4)In the second molar,the pulp stem cells of the inherited permanent teeth were isolated and cultured by enzyme digestion method.The second generation pulp stem cells were selected and stained with alkaline phosphatase and oil red O after osteogenesis and lipogenesis for 21 days.Results:1.The animal model was successfully constructed,and no filling material and root canal post were found to fall off after clinical examination.2.After the modeling is successful,please refer to:(1)Vital signs: in the first and second groups,beagles were in increasing weight,good mental state and normal diet;(2)Clinical manifestations: 4 weeks after the operation in the first group and 8 weeks after the operation in the second group,there were no clinical manifestations such as gingival redness,bleeding and periodontalabscess in the experimental group,glass ionomer fillings were in good condition,no difference with the control group.(3)Mobility test: the first group and the second group of beagles had a mobility of I degree in the experimental group and the control group two weeks after surgery,and the physiological mobility of the same name teeth in the experimental group and the control group was II-III degree four weeks later.The second group of beagles inherited the eruption of permanent canine and molar eight weeks after surgery;(4)He staining: in the first group,there was no inflammatory cell infiltration in the periapical tissue of beagle dogs 4 weeks after operation,but there was no difference between the experimental group and the control group;(5)Hard tissue grinding: the second group of beagle dogs were taken 8weeks after the operation to inherit the permanent canine teeth hard tissue grinding plate.The enamel and dentin structure of the experimental group and the control group were normal;(6)Cell viability test: the pulp stem cells of the experimental group and the control group had strong proliferation ability,ALP and oil red O staining were positive,no abnormality was found.Conclusion:There was no effect of PLA/HA complex on the inheritance of permanent teeth in beagle’s deciduous teeth.
