Objective To investigate the correlation between sex hormone binding globulin and sex hormone binding globulin gene polymorphism(rs6259)of serum lipid and blood glucose in middle-aged and elderly group(age ≥ 45 years)in some urban communities in ningxia. Methods By cluster stratified sampling method,1681 middle-aged and elderly people who met the inclusion and exclusion criteria were investigated in five urban communities in Yinchuan and Wuzhong of Ningxia, including 839 males with an average age of 63.77±10.35 years and 842 females with an average age of 63.28±8.57 years.Genotyping and recording of samples were performed by using MassARRAY system, and statistical software was used to analyze the relationship between SHBG,SHBG SNps polymorphism and blood lipid, blood glucose in different populations. Results 1.The average levels of TC,HDL,LDL and blood glucose in middle-aged and elderly men were lower than those in middle-aged and elderly women(P<0.05),while there was no statistical difference in SHBG levels. After grouping the subjects by gender, the study found that ldl-c and FBG levels increased with age in middle-aged and elderly men, while SHBG levels decreased with age in middle-aged and elderly women(P<0.05).2.Spearman correlation analysis showed a linear negative correlation between SHBG and FBG in the overall study population(r=-0.73 P<0.01).There was a positive linear correlation with age(r= 0.054 P<0.05)and HDL-C(r=0.049 P<0.05).And a positive linear correlation between SHBG level and TC(r=0.218 P<0.05)and TG(r=0.254 P<0.05)in women over 75 years old.Take SHBG as dependent variable, the gender,age,TC,TG,HDL,LDL-C-C,BMI as the independent variable after the multivariate linear regression analysis found no statistical significance.3.With diabetes as the dependent variable, gender, age, abnormal blood lipid level,BMI and hypertension were taken as independent variables for Logistic regression analysis. It can be seen that age and BMI are risk factors for diabetes, while normal TG and blood pressure are protective factors for diabetes.Taking dyslipidemia as the dependent variable and gender, age, body weight,FBG and SH as independent variables,Logistic regression analysis showed that FBG and body weight were risk factors for dyslipidemia. However, there was no significant correlation between SHBG level and diabetes and dyslipidemia.4.There was no significant difference in the distribution of rs6259 genotype frequency, allele frequency, dominant model frequency, invisible model frequency and additive model frequency between men and women in the middle-aged and elderly population of Ningxia Hui Autonomous Region(P > 0.05),and there was no significant difference in BMI,TC,TG,HDL-C,LDL-C,FBC,SHBG among the middle-aged and elderly people without rs6259 genotype.5.In the obese men,the TC level of the men with GG genotype were lower than those with GA or AA(P<0.05).6.Rs6259 dominant model and invisible model had no significant difference in the distribution of dyslipidemia and diabetes. Conclusion The level of SHBG in the middle-aged and elderly population in the urban area of Ningxia increased gradually with the growth of age, among which SHBG and FBG were negatively correlated, while were positively correlated with HDL-C.Levels of TC were lower in obese men with Rs6259 GG genotype, but were not associated with the prevalence of hyperlopoproteinemia. The levels of sex hormone binding globulin and single nucleotide polymorphisms were not found to be associated with type 2 diabetes and hyperlipidemia.
