Objective Using gene methylation chip technology,we screened out gene methylation biomarkers that identify benign and malignant lung tumors.To verify the methylation status of EVX2 and ZNF577 genes in specimen tissues and plasma,and explore the relationship between the methylation status of promoters of this gene and the pathological type,pathological stage,gender,age,smoking,drinking,etc.Methods A total of 60 lung tumor specimens were collected in this study,including 40 non-small cell lung cancer specimens and 20 benign lung specimens.The cancer tissue specimen was the experimental group,and the paracancerous tissue and lung benign nodule tissue were the control group.The following experimental operations were performed:(1)gene methylation chip sequencing,and the target gene was selected;(2)tissue DNA and plasma were extracted separately DNA;(3)Quantitative detection of DNA concentration and purity by UV spectrophotometer,1% agarose gel electrophoresis quality inspection;(4)DNA samples subjected to methylation treatment,2.5% agarose gel electrophoresis on the target gene promoter Methylation status was interpreted;(5)Statistical analysis was performed using spss24.0 software.Results1.The target genes were identified as EVX2 and ZNF577.The difference between the methylation status of the promoter of the target gene in lung cancer tissues and the corresponding adjacent tissues and benign nodule tissues of the lungs was statistically significant(P <0.05).2.The differences between the methylation status of EVX2 and ZNF577 genes in the free DNA of lung cancer plasma and the plasma of benign nodules in lungs were statistically significant(P <0.05).3.The methylation status of EVX2 and ZNF577 gene promoters in lung cancer tissues is consistent with their corresponding plasma methylation status of EVX2 and ZNF577 genes.4.The methylation status of EVX2 and ZNF577 gene promoters in lung cancer tissues were not statistically significant between pathological stage,pathological type,smoking and drinking(P> 0.05)5.The methylation status of EVX2 and ZNF577 genes in lung cancer plasma and smoking were statistically significant(P <0.05),the methylation status of EVX2 gene and drinking alcohol were statistically significant(P <0.05);There was no significant difference in the methylation status of ZNF577 gene with pathological stage and pathological type(P> 0.05).Conclusion1.The methylation status of EVX2 and ZNF577 gene promoters in lung cancer tissues is significantly higher than that in paracancerous tissues and lung benign nodule tissues;2.The methylation status of EVX2 and ZNF577 genes in lung cancer plasma is significantly higher than that in lung benign nodules;3.The methylation of EVX2 and ZNF577 genes in lung cancer tissues is consistent with the methylation of EVX2 and ZNF577 genes in plasma.
