Product Research And Development And Quality Evaluation Of Seabuckthorn Extract

Objective:Sea-buckthorn,as a traditional Chinese medicine with the same origin of medicine and food,contains rich resource component and has high medicinal and edible value.The extract of sea-buckthorn was extracted,concentrated and dried,and the quality of the extract was controlled by the content determination of flavonoids,water and narcissus,and through the network pharmacology study of sea-buckthorn,the components involved in lowering blood lipid in sea-buckthorn were discovered,and the corresponding targets and pathways of these components were analyzed to construct the network diagram of sea-buckthorn's lipidlowering component-target-pathway,and the accuracy of network pharmacology was verified by docking the lipid-lowering components of sea-buckthorn with the key targets.Then the extracts of sea-buckthorn were prepared into dispersible tablets and pressed tablet candies and their quality was evaluated.The above research provides scientific support for the comprehensive utilization of sea-buckthorn resources and the extension of the industrial chain.Method:1.The collected sea-buckthorn was examined according to pharmacopoeia,including appearance,impurity,moisture,ash content,acid-insoluble ash content,total flavonoids,isorrhamidin,etc.Through these tests,it can be determined whether the collected sea-buckthorn meets the requirements of the 2015 pharmacopoeia for sea-buckthorn.2.The extract was extracted from the examined sea-buckthorn,and the extract was concentrated to obtain the extract.The extract was dried by three different drying methods,and the total flavonoids and moisture content of the three drying materials were determined to determine the best drying method.3.Network pharmacology research method is applied to analyze the composition of blood lipid in sea-buckthorn,summarizes related to fall hematic fat composition in the seabuckthorn,targets and pathways,using cytoscape software rendering of sea-buckthorn fall hematic fat ingredient-targets-channel network diagram,using GOLD software will fall hematic fat composition and key target in the sea-buckthorn docking accuracy verification network pharmacology.4.The preparation of sea-buckthorn extract into dispersible tablets and pressed tablets confectionery,the design of a single factor test method to screen the prescription of dispersible tablets and pressed tablets confectionery,and the dispersible tablets and pressed tablets confectionery according to the provisions of pharmacopoeia tablets inspection method for inspection.Result:1.All tests of sea-buckthorn according to pharmacopoeia are in accordance with the provisions of pharmacopoeia and the collected sea-buckthorn can be used for follow-up experiments.3.The sea-buckthorn was extracted by ultrasonic at 80? for 2 times,each time for 2 hours,with 60% ethanol as solvent,feeding at the ratio of 1:8,combined with the extraction solution for decompression and concentration,and freeze-dried to obtain the sea-buckthorn extract.Under this extraction method,the extraction rate of sea-buckthorn could reach 20%-25%.2.Through the network pharmacology study of sea-buckthorn obtain seven components related to fall hematic fat,and these seven elements corresponding to 24 targets,44 pathway,and build the sea-buckthorn fall hematic fat ingredients-targets-access network diagram,and find three key targets through protein-protein interactions.The seven lipid-lowering components in sea-buckthorn were simulated and docked with the three targets,and the pictures of the combination of components and proteins were obtained by using VMD software.4.The sea-buckthorn extract was prepared into dispersible tablets and pressed tablets,and the formula of dispersible tablets was selected by single factor style method.The formula of dispersive tablets was obtained: 35% sea-buckthorn extract,50%MCC,10%PVPP,2% magnesium stearate,3% flavoring agent,and the formula of pressed tablets: 30% sea-buckthorn extract,10% proteinose,15%MCC,43% isomaltitol alcohol,and 2% magnesium stearate.The dispersible tablets of sea-buckthorn extract prepared according to the prescription met the requirements of pharmacopoeia for dispersible tablets,and the dissolution rate was significantly higher than that of ordinary tablets and pressed candy meets the requirements of pharmacopoeia for chews.Conclusion:The dried fruits of sea-buckthorn collected from longde county ningxia province were inspected to meet the requirements of pharmacopoeia.The network pharmacology study of sea-buckthorn found that the components,targets and pathways of lowering blood lipid in sea-buckthorn were basically consistent with the literature reports.The extracts were prepared into dispersible tablets and pressed tablets according to the prescription selected by single factor experiment,All meet the requirements of pharmacopoeia,and this paper provides scientific data for the comprehensive utilization of sea-buckthorn resources and the extension of the industrial chain.
