Objective: Based on the method of network pharmacology,the biological network of Simiao pills in the treatment of gout was constructed and analyzed to explore the effective components and mechanism of Simiao pills in the treatment of gout.Methods: the chemical components of Simiao pills was screened by tcmsp database and ADME as standard,and supplemented by literature;the potential targets of chemical components were predicted by tcmsp database;the targets related to gout were screened by genecards database,the target proteins related to gout were expanded by string database,and the PPI network related to gout was constructed;Cytoscape was used to construct the relationship network of "traditional Chinese Medicine-chemical composition-target-disease",and analyze the topological structure of the network to make clear the effective composition and target of Simiao pills in treating gout;Schrodinger software were used to carry out molecular docking to verify the combination of key chemical composition and core target;In order to explore the mechanism of Simiao pill in the treatment of gout,Go function enrichment analysis and KEGG biological pathway enrichment analysis were carried out by using David database.Results: Simiao pills can treat gout by 16 effective components such as scutellarin,rutaecarpine and berberine acting on 11 targets such as TNF,IL4 and ESR1.According to p< 0.05,19 go functional items were screened.It is speculated that Simiao pills plays an important role in the treatment of gout by participating in immune response,inflammatory response,regulating gene transcription and expression,inducing cytokine activity and nitric oxide biosynthesis,reducing the external apoptotic signal pathway and cell proliferation of ligands.According to p< 0.05,23 KEGG pathways were screened,It is speculated that Simiao pills plays an important role in the treatment of gout by regulating the pathways of glucose and drug metabolism,inflammation and immunity,such as type I diabetes,drug metabolism cytochrome P450,osteoclast differentiation,MAPK,NF KB and toll like receptor.Conclusion: in this study,the effective components,target and biological pathway of Simiao pills in the treatment of gout were systematically analyzed by using the network pharmacology method,which laid a foundation for the study of gout mechanism and the research and development of new anti gout drugs.
