Abstract/Summary:
Objective:To investigate the mechanism of MAPK1 affecting NE secretion through SRF signaling pathway and its role in preeclampsia.Methods:1.Pregnant women who had regular check-up in the second hospital of shanxi medical university from February 2018 to July 2019 were selected as the study subjects.2.They were divided into normal pregnant women group and preeclampsia group,with 30 cases in each group.3.The neutrophils in the peripheral blood of the two groups of pregnant women were extracted,their morphology was observed and counted in primary culture,and then stored in an ultra-low temperature refrigerator at-80℃.4.MAPK1 RNA levels in peripheral blood neutrophils were detected by real-time PCR in the two groups of pregnant women.5.The expression levels of SRF and NE in peripheral blood neutrophils were detected by western blot.6.SPSS22.0 was used for processing,and the experimental results were expressed as mean±standard deviation.Independent sample t test was used for comparison between the two samples,and Pearson correlation analysis was performed between SRF and NE.7.Obtain the approval of the ethics committee of the second hospital of shanxi medical university.Results:1.A comparison of clinical data between the two groups showed that there was no statistical difference in age between the preeclampsia group and the normal pregnant women group[age:(29.18±3.64)、(28.90±3.17)](t=0.36,P>0.05).The pre-pregnancy BMI of the preeclampsia group was significantly higher than that of the normal pregnant women group[pre-pregnancy BMI:(24.91±4.15)、(21.44±2.89)](t=4.35,P<0.05),and the neonatal birth weight of the preeclampsia group was significantly lower than that of the normal pregnant women group[neonatal birth weight:(2512.90±724.53)、(3338.50±400.11)](t=-6.30,P<0.05).2.The number of neutrophils and platelets in peripheral blood of the preeclampsia group was higher than that of the normal pregnant women group[The number of neutrophils:(9.35±2.17)、(7.17±1.25)](t=3.69,P<0.05)[The number of platelets:(226.35±60.49)、(194.67±50.30)](t=2.39,P<0.05).3.Real-time PCR was used to detect the expression level of MAPK1 RNA in peripheral blood neutrophils.The results showed that there was no significant difference in the expression level of RNA between the pre-eclampsia group and the normal pregnant women group[2-??CT:(0.99±0.40)、(0.97±0.03)](t=0.96,P>0.05).4.The expression level of SRF protein in peripheral blood neutrophils was detected by western-blot,and the results showed that the preeclampsia group was significantly up-regulated compared with the normal pregnant women group[gray level:(1439.03±241.63)、(879.31±134.49)](t=4.05,P<0.05)。5.The expression level of NE in peripheral blood neutrophils was detected by western-blot,and the results showed that the pre-eclampsia group was significantly up-regulated compared with the normal pregnant women group[gray level:(1164.65±161.91)、(361.61±34.30)](t=9.70,P<0.05).6.Pearson correlation analysis showed that SRF and NE in neutrophils had a significantly strong positive correlation(r=0.878,P<0.01).Conclusion:There was a significant positive correlation between SRF and NE in neutrophils.It is suggested that SRF can activate neutrophils and increase the level of elastase,leading to vascular endothelial cell injury,which is related to preeclampsia.
