Objective:This experiment by studying the related parameters of oxidative stress in patients with pelvic organ prolapse palace expression,ligament tissue adjacent to clarify in patients with pelvic organ prolapse in ligament tissue adjacent to the palace there is oxidation levels rise,to explore the oxidative stress reaction in pelvic organ prolapse in disease pathogenesis,further provides new treatment for pelvic floor disorder prevention is proposed.Methods:In this study,22 patients admitted to the department of obstetrics and gynecology of the second hospital of shanxi medical university from October 2018 to December 2019 who underwent total laparoscopic hysterectomy and vaginosacral fixation for pelvic organ prolapse(POP?-?)were selected as the experimental group(POP group).Meanwhile,26 patients who underwent transabdominal or laparoscopic total hysterectomy due to benign gynecological diseases instead of pelvic organ prolapse in the department of obstetrics and gynecology of this hospital during the same period were selected as the control group(non-pop group).The main uterine ligament and the sacral ligament tissues of the patients were collected.First,the histopathological features were observed by Masson staining and aldehyde fuchsin staining,and the tissue of the collateral ligament was preliminarily determined.Then,immunohistochemistry was used to detect the expression of 8-OHdG and 4-HNE in the ligament tissues of the patients,and western-blot was further used to detect the expression of 4-HNE and Nrf2.Results:1.The histopathological characteristics of aldehyde-fuchsin staining and Masson staining were observed to further determine the sampling sites: in aldehyde-fuchsin staining,elastin was purplish to dark purple,and compared with non-pop patients,elastin distribution in tissues of POP patients was loose,broken and disordered,and the overall staining was relatively light.In Masson staining,the collagen fibers were red,while the muscle fibers were stained blue-green.In the tissues of non-pop patients,the collagen fibers were neatly and continuously arranged in a wavy shape,while in the tissues of POP patients,the collagen fibers were few in number,irregularly arranged and fragmented,and the overall staining was shallow.2.Immunohistochemistry was used to detect the expression of the indicators related to the oxidation reaction in the non-pop group and the POP group.The results showed that the positive expressions of 8-OHdG and 4-HNE in the sacral ligament and the main ligament in the POP group were significantly higher than those in the non-pop group,and the expressions of the two groups were positively correlated.There was no statistical difference between the two groups.3.Western-blot analysis showed that the relative expression level of 4-HNE in the main ligament of uterus and the sacral ligament in the non-pop group was lower than that in the POP group,and the relative expression level of Nrf2 in the main ligament and the sacral ligament in the non-pop group was higher than that in the POP group,showing a negative correlation.Conclusion:1.High expression of 8-OHdG and 4-HNE in the main ligament and sacral ligament tissues of POP patients suggests that oxidative stress response is involved in the occurrence and development of pelvic organ prolapse.2.The increase of oxidation level leads to the decrease of Nrf2 expression and the activation obstacle of related antioxidant pathway,providing molecular biological mechanism for the occurrence and development of POP.
