Objective:In this study,ifenprodil was applied to treat rats with cerebral ischemia reperfusion injury(IRI),to investigate its effect on NR2 B subunit expression and the effect of brain protection.Methods:Healthy adult male SD rats were randomly divided into 7 groups: 3 ifenprodil groups(ischemia 1h,2h,3h),3 placebo groups(ischemia 1h,2h,3h),and 1 sham group,neurological deficit scorescore was performed on the rats after 24 h reperfusion.After the score,TTC staining was used to measure the necrotic volume of brain tissue,HE staining was used to observe the basic morphologic changes of brain tissue cells,and Immunohistochemistry was used to determine the expression of NR2 B subunit in brain cells.Results:Before 30 min of ischemia for 1h and 2h reperfusion,the application of ifenprodil could effectively decrease the score of nerve function defect score,decrease the infarct volume and decrease the expression of NR2 B in rats,with statistical differences.Compared with the control group,the NR2 B expression was decreased when ifenprodil was given 30 min before 3h reperfusion,showing statistical difference.There was no statistical difference in nerve function defect score and infarction volume between the control group and the control group.Conclusion:Ifenprodil can alleviate cerebral ischemia reperfusion injury after early ischemia reperfusion and play a role in protecting cerebral nerves.The mechanism of ifenprodil in reducing ischemia reperfusion injury may be related to reducing the expression of NR2 B subunit and reducing the toxicity of excitatory amino acids after early ischemia reperfusion.
