Objective:To study the effects of clostridium caseinate and bacillus coagulans on mucous secretion in the mucous layer of the intestinal tract,so as to provide help for the treatment of ulcerative colon.Methods:Zebrafish were taken as experimental animals,and their eggs were collected after natural spawning.Zebrafish eggs were raised in batches in 20mlE3 medium with methylene blue added.The eggs were cultured at 28.5°C until 1 day after fertilization(dpf),and then used E3 medium without methylene blue to 3 dpf.Intervention was added to each group of medium at 3dpf,which was set to normal control group(control group),Clostridium butyricum group(L Group),Bacillus coagulans group(N Group),0.5% DSS(DSS group),0.5% DSS and Clostridium butyricum group(DSS + L Group),0.5% DSS and Bacillus coagulans group(DSS + N Group),and alcian blue staining was used to observe intestinal mucus layer at 6dpf(3 days after exposure)and count,the data for statistical analysis.Results:Clostridium caseate can promote the increase of mucus secretion in zebrafish ulcerative colitis,but bacillus coagulans did not show obvious promoting effect,but it can be seen that the staining point of albino blue becomes larger.When the zebrafish were stimulated by single bacteria only,compared with the control group and N group,the staining count of Alxin blue increased in L group,which was statistically significant(P<0.05),but N group had no significant difference compared with normal control group.The model of zebrafish ulcerative colitis exposed to DSS was treated with Clostridium butyricum(DSS + L Group)and alcian blue staining.Compared with DSS group and DSS N group,the count of alxin blue staining was significantly increased(P<0.05).After intervention with bacillus coagulans(DSS+N group),the alxin blue staining was increased compared with the DSS group and the DSS+L group.Conclusion:Clostridium caseinate can stimulate the intestinal mucus secretion of ulcerative colitis,the bacillus coagulans did not show obvious promoting effect.
