Objective:A rat model of chronic hypoxic pulmonary hypertension was established,to investigate the changes and significance of von will brand factor(vWF),protein C(PC),tissue plasminogen activator(t?PA)and plasminogen activator inhibitor-1(PAI?1)in chronic hypoxic pulmonary hypertension.Methods:1.30 experimental rats were randomly assigned to the normoxic control group,the hypoxic 3-day group,the hypoxic 7-day group,the hypoxic 14-day group and the hypoxic 21-day group,the rat model of pulmonary hypertension was established by chronic hypoxia [oxygen concentration(10.0±0.1)%].2.Right ventricular systolic pressure(RVSP)was measured by right ventricular catheter method in each group,and the right ventricular hypertrophy index(RVHI)was obtained by calculating the weight ratio between the right ventricle(RV)and [left ventricle(LV)+ ventricular septum(S)],i.e.,RV/(LV+S).3.The lung tissues in the same part of each group were collected and stained with HE,VG and Masson,and then pathological changes in pulmonary artery were observed under light microscope.4.Blood was collected from each group of rats through the abdominal aorta,and plasma von Willebrand factor(vWF),protein C(PC),tissue-plasminogen activator(t-PA)and plasminogen activator inhibitor-1(PAI-1)in rats were analyzed by enzyme-linked immune sorbent assay(ELISA).Results:1.Compared with the normoxic control group,RVSP and RVHI were significantly increased in each group under low oxygen environment on the third day,7th day,14 thday and 21 th day(p < 0.01);Pathological examination showed that the pulmonary artery structure changed obviously on the third day,7th day,14 th day and 21 th day.2.vWF,t-PA(except hypoxia group of day 7)and PAI-1 increased significantly after chronic hypoxia when compared with the normoxic control group(p<0.01).Especially,vWF and PAI-1 got peak in the hypoxia group on the 7th day(p<0.01).Compared with the control group,PC decreased obviously after hypoxic exposure on the third day,14 th day and 21 th day(p<0.01),PC increased significantly after hypoxic exposure on the 7th day(p<0.01).3.RVSP and RVHI were positively correlated with the vWF and PAI-1(p<0.01).4.Compared with thenormoxic control group,the ratio of t ? PA to PAI-1 was significantly lower on the third day,7th day,14 th day and 21 th day(p<0.01).Conclusion:1.The rat model of chronic hypoxic pulmonary hypertension(CHPH)was successfully established.2.Changes of coagulation-fibrinolysis factors in rats with chronic hypoxic pulmonary hypertension(CHPH)are obvious,vWF or PAI-1 can be used as an indicator to assess the severity of chronic hypoxic pulmonary hypertension(CHPH).
