| A large number of studies have shown that excessive free radicals can cause oxidative stress damage to the body,trigger a series of diseases and accelerate aging.In the method of tracking free radicals,it is more frontier that the mitochondrial matrix-targeted superoxide indicator circularly permuted yellow fluorescent protein(cpYFP)can display the mitochondrial spontaneous burst superoxide formation in real time.Some researchers studied the free radicals by constructing a C57BL/6-TgH(mito-cpYFP)mouse model,but the lifespan,redox state and behavioral science of C57BL/6-TgH(mito-cpYFP)mouse model is rarely involved.In this study,the expression of cpYFP was measured by RT-qPCR in the main organs of C57BL/6-TgH(mito-cpYFP)mice,Comparing life,behavior,and some biomarkers of major organs in C57BL/6-TgH(mito-cpYFP)mice and wild-type mice.The main results are as follows:(1)The amount of expression from more to less followed by lung tissue,heart tissue,brain tissue,liver tissue,kidney tissue in major organs of C57BL/6-TgH(mito-cpYFP)mice.(2)Compared with wild-type mice,the lifespan and memory of C57BL/6-TgH(mito-cpYFP)mice increased significantly,and there was no significant change in anxiety,learning ability,endurance and balance.(3)Compared with wild-type mice,in liver tissues of C57BL/6-TgH(mito-cpYFP)mice,there was no significant change in the content of NOS,GSH-Px,H2O2 and O2·-,there was significantly decreased in the content of CAT,SOD,MDA and ORP.In the lung tissue,there was significantly increased in the content of MDA,H2O2 and O2·-,there was no significant change in the content of NOS and GSH-Px,there was significantly decreased in the content of CAT,SOD and ORP.In the kidney tissue,there was significantly increased in the content of CAT,NOS and MDA,there was no significant change in the content of SOD,GSH-Px and H2O2,there was significantly decreased in the content of O2·-and ORP.In the brain tissue,there was significantly increased in the content of MDA and H2O2,there was no significant change in the content of NOS,GSH-Px and ORP,there was significantly decreased in the content of CAT,SOD and O2·-.In the heart tissue,there was significantly increased in the content of MDA,H2O2 and ORP,there was no significant change in the content of NOS and GSH-Px,there was significantly decreased in the content of CAT,SOD and O2·-.The above results preliminarily confirmed that the expression of cpYFP in mice can affect the lifespan,behavior and organ redox state of mice,which provides relevant reference materials for the future study of Mito-cpYFP transgenic mice. |
