Background Rectal cancer is one of the most common malignant tumors in the digestive tract,and most of the patients had a late clinical stage when they were diagnosed,so the effect of surgery alone is not good,and surgery combined with other treatment methods become a new direction of rectal cancer treatment.For locally advanced rectal cancer patients,the implementation of preoperative radiochemotherapy become an important means in the clinical work.However,it is different in individual radiosensitivity between patients with rectal cancer,leading to different treatment effect on preoperative radiochemotherapy.So it is an urgent problem to select the patients who are sensitive to radiotherapy by specific molecular marker.A lot of researches confirmed that the micro RNA not only played an important role in the incidence of malignant tumor development and the process of evolution,but also helpd predict tumor diagnosis,treatment and other aspects.With the depth study of micro RNA,it is expected to become a forecast indicator on the tumor diagnosis,predicting clinical curative effect.Multiple studies have demonstrated that mi R-93-5p is closely related to a variety of malignant tumors.Objective The purpose of this study was to investigate the effect of mi R-93-5p expression in rectal cancer cells SW837 on the proliferation and cloning ability and the relationship between mi R-93-5p and radiosensitivity on rectal cancer cell.Methods 1.The expression of mi R-93-5p in rectal cancer cells SW837 was increased by transfection with mi R-93-5p mimic.Four groups were set up: control group;mimic transfection group;radiotherapy group;transfection combined with radiotherapy group.2.The expression of mi R-93-5p in rectal cancer cells was reduced by transfection of SW837 with mi R-93-5p inhibitor.Four groups were set up: control group;inhibitor transfection group;radiotherapy group;inhibitor transfection combined with radiotherapy group.3.The effects of mi R-93-5p on the proliferation and cloning ability of rectal cancer cells and the relationship between mi R-93-5p and radiosensitivity were analyzed by CCK8 and plate cloning experiments.Results 1.After the transfection of mi R-93-5p mimic in rectal cancer cells SW837,the cell proliferation ability was enhanced in the CCK8 experiment and the cell cloning ability was enhanced in the plate cloning experiment.2.After the transfection of mi R-93-5p inhibitor in SW837 rectal cancer cells,the cell proliferation ability was decresed in the CCK8 experiment and decreased in the plate cloning experiment.3.Using 2*2 factorial design variance analysis to analyze the results of CCK8 and plate cloning experiments,it was concluded that there was no significant interaction between mi R-93-5p and radiosensitivity on rectal cancer cells.Conclusion 1.The increased expression of mi R-93-5p can enhanced cell proliferation and cloning ability,while the decreased expression of mi R-93-5p can inhibit cell proliferation and cloning ability.2.There is no significant correlation between mi R-93-5p and radiosensitivity on rectal cancer cells.
