Abstract/Summary:
Objective:In this experiment,a vascularized model was constructed to pre-construct a transplantable pedicled fascia prosthesis complex for ear reconstruction in vivo,To construct a vascularized tissue engineering chamber model for ear reconstruction with pedicled fascial ear prosthesis composite in vivo,and to provide experimental basis for clinical practice by comparing it with traditional methods of implanting ear prosthesis and tissue engineering chamber.Methods: Twenty-four healthy New Zealand white rabbits were randomly divided into three groups.Skin preparation was performed before operation.After satisfactory anesthesia,an experimental model was established.Group A: Simple ear prosthesis was implanted under abdominal wall.Group B: Ear prosthesis was implanted under abdominal wall with tissue engineering chamber.Group C: the superficial abdominal vascular pedicle was elevated and inset into a tissue engineering chamber which had been placed the ear prosthesis.After 4 weeks of operation,the new tissue were analyzed by macroscopic observation,histology,Micro-CT scan and three-dimensional reconstruction,and the morphology and vascularization degree and clinical applicability of the new tissue ear prosthesis complex were evaluated.Results: After 4 weeks,all groups in the experimental could form fibrous connective tissue with different degrees of vascularization and tissue morphology on the surface of the ear prosthesis,which could observe Collagen fibers,neovascularization(red blood cells can be seen inside)and inflammation Cell infiltration.The nascent fibrous connective tissue formed in group C,which was introduced into the superficial abdominal vessel,had a high degree of vascularization.The gross appearance,histology,and Micro-CT scan analysis indicated good vascularization,through vessel density,total vessel volume,and total vessel volume.The total volume analysis of the tissues showed that the blood vessel level was significantly higher than the other two groups(P<0.005).The ratio of total vascular surface area to total vessel volume and mean vessel diameter analysis indicated that the average blood vessel diameter of group B and group C was significantly larger than that of group A,but B.There were no significant differences between the groups and the C groups.At the same time,the results of HE,MASSON and immunohistochemistry showed that the collagen fibers in the new fascia tissue of the new fiber connective tissue in group C were more abundant than the other two groups,and the arrangement was more neat and the inflammatory reaction was lighter.Conclusion: The vascularized pedicled fascia flap ear prosthesis complex can be spontaneously formed in vivo by simply introducing the distally ligated superficial abdominal vessel in the tissue engineering chamber which had been placed the ear prosthesis.
