Objective:To analyze the correlation between the gut microbiota and the rash caused by cetuximab treatment advanced colorectal cancer.The research including the impact of antibiotic use on the incidence of rash and the analysis of intestinal flora in different groups,and to provide theoretical basis for further prevention of the occurrence of rash and possible treatment pathways.Method:1.Retrospective investigation to collect the standard of medical record data of advanced colorectal cancer patients who received cetuximab treatment in top three hospital in Fujian province during 2017.07.01 to 2018.10.20.The rash situation,and other related clinical indicators was tracked and recorded according to the patient information returned to the hospital for review and telephone followup.Some of these patients have collected stool samples.2.The samples were sent to genomic company for 16 S rRNA sequencing and analysis(v3-v4 region).The abundance and diversity of different groups of gut microbiota were analyzed by DNA extraction,PCR amplification,fluorescence quantification,library construction and sequencing.3.SPSS 23.0 software was used for statistical analysis in this study.The clinical features were analyzed using the chi-square test(personc2)or Fisher's exact test.All hypothesis tests were performed using the bilateral test,and the confidence interval was 95%,p<0.05 was considered statistically significant.Results:Total of 38 patients were enrolled,and the incidence of rash was 78.9%.Analysis of the bacterial population showed that the ?-diversity and ?-diversity of the intestinal flora of the rash group were higher than those without the rash.At the phylum level,Firmicutes and Bacteroidetes increased in the rash group,while Proteobacteria increased in the non-rash group.At the genus level,the abundance of Bacteroides in the rash group increased,and in the non-rash group,the abundances of Enterobacter,Mycobacterium,Parabacteroides and Haemophilus were increased.The results of LDA analysis showed that Clostridium,Fusobacterium,Helicobacter,Rumenaceae,and Bacteroides were different genus of rash group,and the Proteobacteria,Enterobacter,and Weirongaceae were the differences of non-rash group.Conclusion:The 16 S rRNA analysis showed that the intestinal flora ?-diversity and ?-diversity of the rash group were higher than that of the non-rash group,and there were specific flora in the two groups.
