Background:Parkinson's disease is a neurodegenerative disease common in middle-aged and elderly people.The incidence rate is 2.0%among the elderly older than 65 years old.The prevalence of PD is also increasing year by year,placing a heavy burden on society and families.Early diagnosis of PD is difficult,and TCS(transcranial ultrasonography)can be used as a Class A recommendation for early diagnosis and differential diagnosis of PD.The pathogenesis of PD is unclear.The SNCA gene is the important pathogenic site of sporadic PD.Multiple polymorphic loci(rs11931074,rs356165,rs356219)have also been confirmed to be associated with the risk of PD.Objective:To study the genotype,sex,age and course of disease in the substantia nigra and echogenic regions(rs11931074,rs356165,rs356219)of PD patients in the Han population of Fujian,China,and to investigate the SNCA gene polymorphism and Correlation of substantia nigra ultrasound in Parkinson's disease.Methods:A total of 276 patients with PD who were admitted to the Department of Neurology,Union Hospital of Fujian Medical University from February 2017 to May2018 were included in the study.All the patients underwent transcranial ultrasonography.The high echogenic area of the substantia nigra was used as the basis for the judgment.The black matter positive group and the substantia nigra negative group were divided into 0.2cm~2.SNCA locus was detected in 276 patients by sanger gene sequencing technology.The detected gene loci include rs11931074,rs356165and rs356219.The correlation between SNCA gene polymorphism and substantia nigra ultrasound in Parkinson's disease was analyzed by two-category logistic regression.Results:The alleles of rs11931074,rs356165 and rs356219 were not significantly different between the substantia nigra positive group and the substantia nigra negative group in the PD group.rs356165 and rs356219 may be positively correlated with the substantia nigra hyperechoic of PD patients in the Han population of Fujian,China.Rs11931074 is negatively correlated with substantia nigra.In this group of PD patients,the genetic variation occurring at rs11931074 was negatively correlated with substantia nigra ultrasound for age>60 years;for age>60 years,disease duration?5years,genetic variation and black occurred at rs356165 Quality ultrasound was positively correlated;for female,age>60 years,duration?5 years,genetic variation at rs356219 was positively correlated with substantia nigra.Conclusion:SNCA gene polymorphisms rs11931074,rs356165 and rs356219 are associated with substantia nigra ultrasound in PD patients with Han population in Fujian,China.
