γδT cells are an important subset of T lymphocytes,and γδT cells can be divided into two major subgroups based on the difference of TCR chain structures,namely,Vδ1 and Vδ2.Vδ2 cell subsets have the characteristics of anti-infection and anti-tumor.And Vδ2 subset only exists in humans and primates.Researches showed that Vδ2 T cells can inhibit the growth and metastasis of tumors.Vδ2 T cells could be potentially developed into promising immunological therapeutics against tumor.Selenium is one of the essential trace elements in the human body and plays very important biological functions,such as anti-tumor,anti-oxidation,and antagonism of heavy metals,etc.However,inorganic selenium has strong toxicity and it’s not easily absorbed.The single-state nano-selenium particles have promising biocompatibility,and can be applied as anti-oxidants,antibacterial agent and anti-tumor etc.Currently,as one type of bionanomaterials,anti-tumor applications of nano-selenium have been greatly appreciated.However,only a few reports have shown that nano-selenium can upregulate the immune function.The purpose of this work is to study the regulation effect of nano-selenium on human T cell function.Our study found that nano-selenium can significantly up-regulate the expression levels of several genes associated with proliferation and differentiation of γδ T cells,including xxx,and nano-selenium can enhance the anti-tumor ability of γδ T cells.The expression levels of ICAM-1 and CD16 that are closely related to the killing capacity were significantly upregulated.Furthermore,we found that glycolysis andaerobic respiration were also significantly elevated and cytoskeleton tubulin was upregulated as well.In vitro killing experiments demonstrated that nano-selenium strikingly potentiated γδT cells killing of tumor cell lines such as MCF-7,K562,and SKBR3.Together,our results demonstrate that nano-selenium can upregulate the function of human γδ T cells,particularly can enhance the killing ability of γδ T cells against tumor cells.
