Objective Aseptic loosening of the artificial joint prosthesis is one of the main reasons for the failure of artificial total knee arthroplasty.By detecting related signal molecules in the Notch signaling pathway,the boundary membrane tissue around the artificial knee prosthesis and knee osteoarthritis slip.To investigate the expression of membrane tissues,to explore the role and significance of Notch signaling pathway in the pathogenesis of aseptic loosening of artificial knee joints.Methods In this experiment,21 cases of aseptic loosening of the limiting membrane after total knee arthroplasty were collected as the experimental group,and 30 cases of primary knee osteoarthritis underwent artificial total knee arthroplasty as control.Paraffin sections were made,the degree of aseptic inflammation was observed by HE staining,the cell components were observed by TRAP staining,and the expression levels of Notch1,Jagged1,Hes1,and Hes5 were determined by immunohistochemical SABC method.Use Image pro-Plus6.0 to detect the integrated optical density(IOD)of the expression of related signal molecules,and use the average integrated optical density value to represent the particle density of related proteins.The obtained data were statistically analyzed using SPSS22.0.Results 1.In HE staining,the inflammatory response of the limiting membrane tissue in the aseptic loosening group was obvious,and a large number of black abrasion particles were seen,and more macrophages were infiltrated.In the osteoarthritis group,the inflammatory response of the synovial tissue is relatively light,and the number of inflammatory cells is relatively small;2.In the TRAP staining results,the osteoclasts were found in the boundary tissue of the aseptic loosening group and were distributed on the edge of the boundary membrane.In the osteoarthritis group,there were more light blue fibroblasts.3.The results of immunohistochemistry showed that Notch1,Jagged1,Hes1 and Hes5 were significantly higher than those in the osteoarthritis group in the aseptic loosening group(P <0.05).Conclusion Under the action of abrasive particles,the activation of the Notch signaling pathway causes macrophages to release a large number of inflammatory factors,and then osteoclasts proliferate and activate,promoting osteolysis around the prosthesis.Involved in the mechanism of aseptic loosening after total knee arthroplasty.
