MiRNA-214 Expression In Human Gastric Cancer: Correlation With Disease Progression And Patient Outcome

Objective To investigate the expression level of miRNA-214 in gastric cancer and its relationship between clinicopathological parameters of gastric cancer and patient prognosis.Methods According to the criteria of inclusion and exclusion gastric cancer tissues and paracancerous tissues of 90 matched patients who were pathologically diagnosed after surgical resection were selected to make tissue chips from February 2008 to August 2008.The clinicopathological data and follow-up data of all patients were collected.The expression level of miRNA-214 in gastric cancer and paracancerous tissues was detected by in situ hybridization and combined with clinicopathological data by statistically analyzed.Kaplan-Meier survival analysis and Cox survival regression analysis were used to study the correlation between the expression of miRNA-214 and the prognosis of patients with gastric cancer.Results1.Among the 90 patients with gastric cancer,including 61 males and 29 females.The median age was 65.5 years old and the average age was(64.3 ±10.2)years old.The most common pathological type were adenocarcinoma(47 cases).The most common sites were gastric antrum(35 cases)and lesser curvature(25 cases).After follow-up 58 died cases and 32 survival cases.The average survival time was(71.8 ±25.6)months.2.In gastric cancer tissues has 52 high expression cases and 30 low expression cases of miRNA-214.In paracancerous tissues has 38 high expression cases and 44 low expression cases.Statistical analysis showed that the high expression rate of miRNA-214 in gastriccancer tissues was(64.3%(52/82),which was significantly higher than paracancerous tissues(46.3%(38/82)),and the difference was statistically significant(P=0.028).3.It was found that high expression of miRNA-214 and pathological grade of gastric cancer patients(P=0.550),T stage(P=0.615),M stage(P=1.000),gender(P=0.343),age(P=0.344)had no significant correlation,and the difference was not statistically significant.But it was significantly related to the TNM stage(P=0.002)and N stage(P<0.001)of gastric cancer patients,and the difference was statistically significant.4.It was found that the overall survival rate of gastric cancer patients in the miRNA-214 low expression group was higher than that in the miRNA-214 high expression group,and the difference was statistically significant(P<0.001).5.It was found that the overall survival rate of gastric cancer patients in the miRNA-214low-expression group was higher than that of the miRNA-214 high-expression group in gastric cancer patients' TNM stage I-II,and the difference was statistically significant(P=0.009).The overall survival rate of gastric cancer patients in the miRNA-214 low expression group in the TNM stage III-IV was not related to GC patients in the miRNA-214 high expression group,and the difference was not statistically significant(P=0.213).6.It showed that the factors affect overall survival of gastric cancer patients has pathological grade(P<0.001),T stage(P=0.009),TNM stage(P<0.001),N stage(P<0.001),miRNA-214 expression(P<0.001).Further analysis showed pathological grade(P=0.001)and miRNA-214 expression(P=0.043)is an independent prognostic factor to affect gastric cancer patients.Conclusion1.MiRNA-214 is highly expressed in gastric cancer tissues.2.The high expression of miRNA-214 is related to the N stage and TNM stage of gastric cancer patients.3.MiRNA-214 expression may be a potential biomarker for predicting the prognosis ofgastric cancer patients.
