Objective:To compare the effects of HDL from heathy subjects(HDLHeathy)and HDL from the patients with coronary heart disease(HDLCAD)on the lipid deposition and apoptosis in mouse peritoneal macrophages.Methods:1.HDL was isolated from healthy subjects(HDLHealthy),stable CAD patients(HDLSCAD)and acute myocardial infarction patients(HDLAMI).To observe their effects on the lipid deposition and apoptosis by ox-LDL induced foam cells2.The accumulation of intracellular lipids was determined by oil red O staining.3.The apoptosis of macrophage was measured by fluorescence microscopy with annexin-V/PI staining.4.DCHF-DA,a redox-sensitive dye,was used to detect intracellular ROS levels.5.The protein expression of ABCA-1,ABCG-1,Bcl-2 and Bax was determined by Western blot analysis.Results:1.Lipid deposition in the macrophages was increased significantly after ox-LDL treatment,and the expression of ABCA-1 and ABCG-1 were up-regulated?P<0.05?.2.Compared with ox-LDL treatment alone,HDLhealthy decreased lipid deposition in the macrophages and up-regulating the expression of ABCA-1 and ABCG-1?P<0.05?,lipid deposition in the macrophages was further increased after treatment with HDLSCADCAD and HDLAMI,the expression of ABCA-1 and ABCG-1 were down-regulated?P<0.05?.3.Compared with HDLSCAD treatment.Lipid deposition in the macrophages was further increased after treatment HDLAMI,the expression of ABCA-1 and ABCG-1 was down-regulated?P<0.05?.4.ox-LDL increased the levels of intracellular ROS and apoptosis by reducing the levels of antiapoptotic protein Bcl-2 and inducing the expression of proapoptotic protein Bax.5.HDLHealthy decreased the levels of intracellular ROS and apoptosis by increasing the levels of antiapoptotic protein Bcl-2 and reducing the expression of proapoptotic protein Bax.In contrast,HDLSCADCAD and HDLAMIMI had opposite effects on the intracellular ROS,the macrophages apoptosis and the expression of apoptosis related proteins Bcl-2 and Bax.Conclusion:1.HDLhealthyealthy decreased lipid accumulation in macrophages and decreased macrophage apoptosis.2.HDLCADAD promotes lipid accumulation in macrophages and induces macrophage apoptosis.
