Analysis And Identification Of The Chemical Components In Gualou Xiebai Banxia Decoctionby UPLC-LTQ-Orbitrap-MS

Gualou XiebaiBanxia decoction(GLXB)is origint from JinGuiyaolue which written by ZhongJing zhang at Han dynasty.It contains four Chinese herbal medicine such as Gua Lou,Xie Bai,Ban Xia,Mi Jiu.Gulou Xiebai Banxia decoction,a well-known classic traditional Chinese medicine prescription,has been widely used to treat coronary heart diseases for thousands of years in Eastern Asian countries due to its remarkable clinical effect.Objective:In this study,UPLC-ESI-LTQ-Orbitrap-MS was used to analyze the chemical constituents in GLXB.Method: Firstly,analyze and identify the chemical constituents of single herbs.On the basis of this,the chemical composition of the GLXB and the body samples’(blood,urine,fecal)component and metabolic components were analyzed and identified.The chemical compounds were identified by the accurate mass measurement of molecular ions and fragment ion and comparation with reference substance.Result:UPLC-ESI-LTQ-Orbitrap-MS analysis method was established,and 90 chemical constituents were identified from Gua Lou Fruit;28 kinds of steroid saponin were identified from XieBai;39 chemical components were identified from Ban Xia;Establish UPLC-ESI-LTQ-Orbitrap-MS analysis method,and identify 88 chemical components from GLXB;29 prototype components and 12 metabolic components from plasma,urine and feces samples.Conclusion: This study lay the foundation for the analysis of the pharmacodynamic basis,the quality control research and drug development of the GLXB.
