Objective:To investigate the expression of ACTN-4 in Oral Squamous Cell Carcinoma and the effect of inhibiting of its expression on the proliferation and apoptosis of cancer cells.Methods:qPCR was used to detect the mRNA expression of ACTN-4 in Oral Squamous Cell Carcinoma tissues;Immunohistochemistry was used to detect the expression of ACTN-4 protein in Oral Squamous Cell Carcinoma tissues;SCC15 cells were divided into blank group(without special treatment),NC group(cells were transfected lipofectamine 2000 and NC siRNA)and si-ACTN-4 group(cells were transfected lipofectamine 2000 and ACTN-4-specific siRNA).The expression of ACTN-4,Bcl-2,Bax,AKT and p-AKT was detected by Western blotting,the apoptotic rate was detected by flow cytometry,and the cell proliferation was detected by MTT at 24 h,48h and 72 h after siRNA transfection.Results:(1)The mRNA expression of ACTN-4 in Oral Squamous Cell Carcinoma was significantly higher than that in adjacent tissues(P<0.05).(2)ACTN-4 was localized in the cytoplasm or membrane of oral cancer cells.The positive cells in the cytoplasm or membrane showed obvious yellow or brown color,and the positive cells showed diffuse distribution.(3)si-ACTN-4 was transfected into SCC15 cells for 48 hours,the expression of ACTN-4 protein in the si-ACTN-4 group was significantly lower than that in the blank group(P < 0.05).(4)The OD value in SCC15 cells transfected with si-ACTN-4 for 24 hours was not significantly different from that of the blank group(P>0.05).The OD value in SCC15 cells transfected with si-ACTN-4 for 48 and 72 hours were significantly different from that of the blank group(P < 0.05).(5)The apoptotic rate of SCC15 cells transfected with si-ACTN-4 for 48 hours was significantly higher in si-ACTN-4 group than in blank group(P < 0.05).(6)si-ACTN-4 was transfected into SCC15 cells for 48 hours,compared with the blank group,the expression of p-AKT and Bcl-2 in the si-ACTN-4 group decreased significantly,and the expression of Bax increased significantly(P<0.05).There was no significant difference in the expression of AKT among the three groups(P>0.05).Conclusion:ACTN-4 is positive in human Oral Squamous Cell Carcinoma.Inhibition of ACTN-4 expression can significantly reduce the proliferation of oral squamous cell carcinoma SCC15 cells,induce apoptosis,down-regulate the expression of p-AKT and Bcl-2,and up-regulate the expression of Bax.Inhibiting the expression of ACTN-4 may inhibit the proliferation of SCC15 cells and induce apoptosis by inhibiting the activation of AKT signaling pathway,it may be a potential target for Oral Squamous Cell Carcinoma targeted therapy.
