PART ? Determination of TWEAK and NF-?B levels in lung tissue of asthmatic mice?Objective?To investigate the changes and significance of TWEAK and NF-?B in lung tissue of mice with asthma.?Method?Male Balb/c mice(6 to 8 weeks old)were randomly divided into three groups : normal control group,asthma group and dexamethasone treatment group.Mice in the asthma group were sensitized and challenged with OVA to establish asthmatic model,while mice in the dexamethasone treatment group was treated with dexamethasone prior to each challenge.Within 24 hours after the final OVA challenge,The severity of inflammation in lung tissue was evaluated by hematoxylin and eosin(H&E)staining.The pathological changes were observed under light microscope,the expression and distribution of TWEAK and NF-?B in lung tissue of mice in each group were detected by immunohistochemistry,and the protein of TWEAK and NF-?B in lung tissue of mice in each group was detected by Western-Blot.? Result ?HE staining showed that in the normal control group,the epithelial cells of trachea mucosa were arranged regularly,the morphology was complete,there was no obvious infiltration of inflammatory cells in the submucous membrane,the structure of trachea lumen was normal,and the wall of trachea was smooth and not narrow.In asthma group,airway epithelial cells were disordered and exfoliated,inflammatory cells appeared in submucous and alveolar stroma,especially around blood vessels,trachea wall thickened,alveolar stroma and airway smooth muscle cells proliferated.In dexamethasone intervention group,the epithelial cells of trachea mucosa decreased,less he infiltration of inflammatory cells in submucous and alveolar stroma,the thickening of tube wall was alleviated,and there was no obvious proliferation of alveolar stroma and airway smooth muscle cells.The expression of TWEAK and NF-?B in the group of asthma and dexamethasone was significantly higher than that in the control group(P <0.05).The expression of TWEAK and NF-?B in the dexamethasone intervention group was lower than that in the control group(P <0.05).?Conclusion?TWEAK and NF-?B levels increased in lung tissue of mouse asthma model,and TWEAK and NF-?B were associated with asthma.PART ?Determination of plasma TWEAK concentration and NF-?B activity in children with asthma?Objective?To investigate the changes and significance of TWEAK level and NF-?B activity in peripheral blood of children with asthma.?Method?In the experiment,32 children with asthma were included as the study group.The plasma TWEAK concentration and the activity of NF-?B in peripheral blood mononuclear cells(PBMC)were measured by ELISA method in the attack and remission stages of asthma,and 20 healthy children were used as the control group.? Result ?(1)The level of plasma TWEAK in asthma attack group was higher than that in remission group and healthy control group.(P <0.05);(2)There was no significant difference in TWEAK concentration between asthma remission group and healthy control group.(P >0.05);(3)The activity of NF-?B in PBMC of asthma attack group was higher than that of remission group and healthy control group(P <0.05);(4)The activity of NF-?B in PBMC of asthma remission group was higher than that of healthy control group(P <0.05).?Conclusion?TWEAK and NF-?B were associated with asthma attack in children.TWEAK was a cytokine associated with asthma...
