Objective: To explore the effect of Programmed cell death factor 4 on incidence,clinical characteristics and prognosis of bladder cancer.Methods: From June 2014 to December 2014,72 patients with bladder cancer were collected in the Department of Urology of Hospital.By paired samples collected each patient with bladder cancer tissues and adjacent normal tissues(tumor distance = 2cm).All subjects were followed up regularly.The median follow-up time was 31.2months.Trizol method was used to extract mRNA from bladder cancer tissues and adjacent tissues,and the relative expression level of PDCD4 mRNA in tissues was detected by RT-PCR.Meanwhile,immunohistochemistry was used to detect the expression of PDCD4 protein in the tissues.SPSS21 makes statistical analysis of the data.Results: 1.The positive rate of PDCD4 protein in para cancerous tissues was 59.7%,the positive rate in bladder cancer tissue was 23.6%,and the positive rate of PDCD4 protein in bladder cancer tissue was significantly lower than that of the para cancerous tissue.The relative expression of PDCD4 mRNA in bladder cancer tissues and para cancerous tissues was 0.65 and 1.09 respectively.The relative expression of PDCD4 mRNA in para cancerous tissue was significantly higher than that of bladder cancer tissues.2.Patients with less than 50 years old,pathological grade I,pathological stage is Tis~T1,tumor size is less than 3cm and no recurrence,the positive rate of PDCD4 protein is higher.The relative expression level of PDCD4 mRNA was significantly higher in patients less than 50 years old,grade I and II of pathological grade,metastasis,pathological stage of Tis~T1,tumor size less than 3cm and non recurrence.3.The median survival time of the PDCD4 positive patients was 28.5 months,the median survival time of the PDCD4 negative patients was 19.6 months,and the survival time of the bladder cancer patients with PDCD4 protein positive was prolonged significantly.Compared to PDCD4-negative bladder cancer patients(11.6months),PDCD4 positive bladder cancer patients(20.5 months)had a significantly longer progression free survival time.The median survival time of patients with high expression of PDCD4 mRNA was 33.4 months,and the median survival time of patients with PDCD mRNA low expression was 19.9 months,and the survival time of bladder cancer patients with high expression of PDCD4 mRNA was significantly prolonged.In patients with bladder cancer(11.5 months)with low expression of PDCD4 mRNA(11.5 months),the progression free survival time of patients with PDCD4 mRNA high expression of bladder cancer(20 months)was also significantly prolonged.Conclusion: 1.The relative expression of PDCD4 mRNA and the positive rate of PDCD4 protein in bladder cancer tissues is significantly lower than that in the para cancerous tissue,suggesting that PDCD4 is associated with the occurrence of bladder cancer.2.PDCD4 is associated with the clinical features of age,pathological grading,metastasis,pathological staging,tumor size and recurrence of bladder cancer,suggesting that PDCD4 is associated with the development of bladder cancer.3.The elevated expression levels of PDCD4 mRNA or the positive of PDCD4 protein can significantly prolong the total and progression free survival of the patients,suggesting that PDCD4 is a protective factor for the prognosis of bladder cancer.
