Objective: Gastric cancer is still a major aggressive malignancy worldwide,and is one of the most common malignant tumor in China.Until now,the mechanism of gastric cancer development is still unclear.While the importance of circular RNAs?circ RNAs?involved in carcinogenesis has gradually been acknowledged,their role in human cancers is not largely understood,including in GC.In this study,circ RNA microaary found that hsacirc0001368 was extensively downregulated in gastric cancer tissues.However its expression and function have not been investigated in gastric cancer.At first this study investigated the correlation between hsacirc0001368 expression and development of gastric cancer,and analysised the relationship between hsacirc0001368 expression and clinicopathological parameters or prognosis.Secondly,through molecular biological experiments in vivo and in vitro,we investigated the relationship between hsacirc0001368 expression and biological behaviour of gastric cancer.Finally,we studied the mechanism of hsacirc0001368 affecting the development of gastric cancer.This study aims to investigate the role of hsacirc0001368 and the underlying molecular mechanisms in gastric cancer and hopefully provide a new theoretical basis for potential therapeutic targets of gastric cancer.Methods: 1.Using circ RNA microaary to identify the differentially expressed circ RNA in gastric cancer and Using q RT-PCR to verify.2.Through CCK8 method discussing the relationship between hsacirc0001368 expression and gastric cancer cell proliferation.3.Through Transwell little room investigating the correlation between hsacirc0001368 gene expression and the invasion of gastric cancer cells.4.By building the nude mice subcutaneously into tumor model,validating the influence of hsacirc0001368 in HGC turn-27 gastric cancer cell lines forming subcutaneous tumor in nude mice;5.Using bioinformatics analysis,dual-luciferase reporter gene assay and fluorescence in situ hybridization to determine the mi RNA and its downstream target genes that can bind to hsacirc0001368,and revealing the molecular mechanism of hsacirc0001368 promoting the proliferation and invasion of gastric cancer.Results: 1.Hsacirc0001368 expression levels in gastric cancer tissues were significantly lower than that in normal tissues.2.Hsacirc0001368 can inhibited the proliferation of gastric cancer cells.3.Hsacirc0001368 can inhibited the invation of gastric cancer cells.4.Hsacirc0001368 inhibited subcutaneous tumorigenesis in nude mice.5.Circular RNA hsacirc0001368 suppresses the progression of gastric cancer by regulating mi R-6506-5p/FOXO3 axis.Conclusion: 1.Hsacirc0001368 expression levels in gastric cancer tissues were significantly lower than that in normal tissues;2.Hsacirc0001368 can inhibited the proliferation and invation of gastric cancer cells;3.Circular RNA hsacirc0001368 suppresses the progression of gastric cancer by regulating mi R-6506-5p/FOXO3 axis.
