Mechanism Of LINC00460 Promoting Proliferation Of Gastric Cancer Cells

Background:As one of the most commom malignant tumors,Gastric cancer(GC)ranks the third leading cause of cancer mortality worldwide.Due to the lack of sensitive diagnostic markers,most GC cases were diagnosed at advanced stages with a poor clinical outcomes.Long non-coding RNAs(lncRNAs)are non–protein coding transcripts longer than 200 nucleotides.They can regulate a variety of cell biology processes like proliferation,metastasis,apoptosis,and cell cycle,etc.At present,the research on lncRNAs in GC still remains in infancy period.Exploring the role of lncRNAs in GC can provide a theoretical basis for clinical diagnosis and treatment of GC.Objective:To explore the expression characteristics and functions of LINC00460 in gastric cancer cells,to reveal the significance of LINC00460 in the clinical diagnosis and treatment of gastric cancer.Methods : Bioinformatics analysis was used to screen lncRNAs expressed differentlybetween cancer and normal tissue samples of TCGA gastric cancer database.qRT-PCR technique verified that LINC00460 was highly expressed in GC tissues and cells.The biological function of LINC00460 in GC cells was investigated through colony formation,flow apoptosis and cell cycle assay after LINC00460 knockdown.The effect of LINC00460 on GC cells in vivo was explored through xenograft tumor models in nude mice.Transcriptome microarray analysis and qRT-PCR technique were used to screen the downstream genes of LINC00460 and clarify the regulation function of LINC00460 to E2F1.Results:LINCOO460 was identified highly expressed in gastric cancer through TCGA databaseand qRT-PCR technique.LINC00460 knockdown inhibited proliferation of GC cells and promoted apoptosis and cell cycle arrest in G0/G1 phase.Xenograft mouse model confirmed that LINC00460 knockdown inhibited GC cells' tumorigenic ability in vivo.Transcriptome microarray analysis indicated that LINC00460 increased the expression of E2F1.qRT-PCRtechnique validated the result of transcriptome microarray analysis.Conclusion:LINC00460 is expressed highly in GC tissues and cell lines.LINC00460 promotes the proliferation of GC cells through up-regulating the expression of E2F1 in vitro and in vivo.
