Background: To explore the efficacy of olive oil in the prevention and treatment of postmenopausal osteoporosis(PMOP)in women.The SD rat osteoporosis model was constructed in the experiment.The correlation between the two bone metabolism markers and bone mineral density was studied by detecting the dynamic changes in ?-isomerized forms of type I collagen breakdown products(?-CTx)and procollagen type I N propeptide(PINP)and bone mineral density(BMD),to discuss the changes of bone metabolism markers in the process of osteoporosis and the response after olive oil intervention,to provide experimental basis for evaluating the effectiveness of olive oil,to discuss whether olive oil has potential application in preventing postmenopausal osteoporosis.Methods: 90 female SD rats aged 4 months were randomly divided into three groups: sham operation group(Sham group),ovariectomy group(OVX group),ovariectomy + olive oil group(OVX+Olive group).All rats in the OVX group and the OVX+Olive group were resected bilateral ovaries to construct an ovariectomized SD rat model;In the sham group,only a small piece of retroperitoneal fat similar to the size of the ovary was removed.The rats were administered after 1 week of adaptive feeding: Sham group and OVX group were administered with normal saline at 1 ml/100 g body weight;OVX+Olive group was administered with high quality virgin olive oil at 1 ml/100 g body.All rats were given intragastric administration once a day for 24 weeks.After 8,16 and 24 weeks of administration,10 rats were randomly selected from each group.One week of adaptive feeding,left ventricular blood was taken,and serum samples were taken after centrifugation.The serum concentration of bone metabolism marker PINP and ?-CTx were detected by ELISA kit;and bone mineral density of lumbar vertebrae and right femur in each group were measured by dual energy X-ray absorptiometry.Results: One rat in Sham group and one in OVX group died on the second day after operation.The reason may be that the drug was directly injected into the blood vessel during intraperitoneal injection of anesthetics,resulting in excessive blood concentration and respiratory depression.Compared with the Sham group,the serum concentration of PINP in the OVX group was significantly decreased from the time of surgery to 24 th week after surgery(P<0.05).Compared with the OVX group,the serum concentration of PINP in the OVX+Olive group was significantly increased from 16 th week to 24 th week after surgery(P<0.05).In contrast,compared with the Sham group,the ?-CTx serum concentration of the OVX group was significantly increased from the time of surgery to 24 th week after surgery(P<0.05);compared with the OVX group,the ?-CTx serum concentration in the OVX+Olive group was significantly decreased at the 24 th week after surgery(P<0.05).At the same time,the lumbar spine bone mineral density of the three groups of rats did not differ significantly from the time of surgery to the 24 th week after surgery.However,compared with the Sham group,the femoral bone density of the OVX group was significantly lower at the 24 th week after surgery(P<0.05).Compared with the OVX group,the femoral bone density of the OVX+Olive group was significantly increased at the 24 th week after surgery(P<0.05).Then,we analyzed the correlation between the changes of serum PINP and ?-CTx concentrations and bone mineral density in the later stage of femur.It was found that the absolute changes of serum PINP(P = 0.0049)and ?-CTx(P = 0.0385)levels OVX group were significantly correlated with the absolute changes of bone mineral density of femur compared with Sham group.At the same time,the absolute changes of serum PINP(P = 0.0083)and ?-CTx(P = 0.0276)in OVX+Olive group were significantly correlated with the absolute changes of bone mineral density of femur.However,there was no significant correlation between the absolute changes of serum PINP(P = 0.1158)and ?-CTx(P = 0.2166)and bone mineral density of femur in OVX+Olive group compared with OVX group.Conclusion: In this study,the concentration of PINP and ?-CTx in the blood of SD rats receiving bilateral ovariectomy and olive oil gavage and changes in bone mineral density suggest that high-quality virgin olive oil is intensively consumed for rat bone after ovariectomy.The qualitative effects and the correlation between the changes in the concentration of PINP and ?-CTx and the changes in bone mineral density in rats demonstrate that virgin olive oil has the potential to be used for clinical prevention and treatment of PMOP and to reduce the risk of fracture.
