Objective: This study aims to detect the expression of CD44v6 and hyaluronic acid(HA)in colorectal cancer(CRC),to investigate the effect of CD44v6 and HA on cisplatin resistance and to make a preliminary explore of the mechanism of cisplatin resistance.Methods: A total of 66 tissue specimens diagnosed with CRC by pathology were collected in this study.The expression of CD44v6 was detected by qRT-PCR,western blot and immunohistochemistry.The correlation between the expression of CD44v6 and the clinicopathological characteristics of patients was analyzed.At the same time,the CRC cell lines were cultured,and the expression of CD44v6 was detected in the cell lines by qRT-PCR and western blot.In the study,human hyaluronic acid enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of HA.Using lentivirus to transfect the CRC cell lines to knock down the CD44v6 gene to verify the relationship between CD44v6 and CRC cisplatin resistance.After digesting the CRC cell line with HA enzyme to block the binding of CD44v6 and HA,to explore the relationship between the CD44v6-HA complex and CRC cisplatin resistance.The expression of PI3 K and AKT protein,which play a key role in PI3K/AKT signaling pathway,was detected by western blot on CRC cell lines transfected with lentivirus and treated with HA enzyme.Rusults: For normal colorectal mucosa tissues,the expression of CD44v6 in CRC significantly increased(p ? 0.001).The expression of CD44v6 was statistically different from that of serum CEA(p ? 0.001),and it was not statistically different from the patient's age,gender,TNM staging,tumor differentiation,and tumor site(p?0.05).HA secreted in the CRC cell line RKO(p?0.001).For normal colorectal epithelial cell,the expression of CD44v6 significantly increased in colorectal cancer cell line(p ? 0.001).The cisplatin resistance of CRC cell lines with CD44v6 gene knockdown and HA enzyme treated decreased significantly(p ? 0.05).At the same time,the expression of PI3 K and AKT protein in the PI3K/AKT signal pathway decreased significantly(p?0.01).Conclusion: CD44v6 highly expresses in CRC.The expression of CD44v6 is correlated with serum CEA level.The low expression of CD44v6 reduces the cisplatin resistance of CRC cell lines.Blocking the binding of CD44v6 to HA can reduce the cisplatin resistance of CRC cell lines.The expression of CD44v6 and its combination with HA are involved in the process of PI3K/AKT signaling pathway-mediated CRC resistance to cisplatin chemotherapy.
