Dendrobium officinale Kimura et Migo is a valuable medicinal plant.But Dendrobium officinale and some food and drug homology Chinese medicine together,make compound preparation,can play the more function of Dendrobium officinale.In this study,Dendrobium officinale and several food and drug homologous plants were used to prepare the Dendrobium officinale formula.The effects of Dendrobium officinale formula on lung function,immune function and intestinal flora of C57BL/6 asthma mice were studied by inoculating Dendrobium officinale formula,and licorice powder was used as positive control.The results are as follows:1.Effects of Dendrobium officinale formula on lung function in asthmatic mice: Ovalbumin(OVA)induced asthma mice were improved after oral administration of Dendrobium officinale formula 35 day.In body mass there was no significant difference between mice with asthma and mice with Dendrobium officinale formula.Model group compared to normal mice immunity organ quality and viscera than lower(p<0.001),while Dendrobium officinale formula quality of immune organs than gavaging group compared with model group was increased,but the difference was not statistically significant.Dendrobium officinale formula can make asthma mice immune organ index to rise,but not obvious;From Dendrobium officinale formula in mice bronebial alveolar lavage fluld super oxide dismutase(SOD),catalase(CAT)and total antioxidant capacity(T-AOC)in the analysis,the influence of the Dendrobium officinale formula can improve asthma mice,SOD,CAT,T-AOC level and contrast found that SOD and T-AOC level middle dose group of the most obvious,compared with model group,SOD and T-AOC increased by 68.34% and 101.08% respectively.The improvement of CAT activity was the best in the high dose group,with an increase of 58.64%,indicating that the Dendrobium officinale formula could improve the antioxidant capacity of mice lung tissues and maintain the dynamic balance between the generation and clearance of oxygen free radicals.Classificationand counting of bronebial alveolar lavage fluld inflammatory cells in mice and observe the pathological lung tissue in mice found that effect is most obvious in middle dose group,the total middle dose group of cells,the number of eosinophils,lymphocytes and neutrophils count compared with model group fell by 43%,68.49%,51.75% and 60.00%,respectively,at the same time the middle and high dose group mice lung inflammatory cell infiltration,epithelial cell necrosis falls off phenomenon,bronchial lumen is complete,that Dendrobium officinale formula can improve asthma airway inflammation in mice,improve lung function in mice.2.Effects of Dendrobium officinale formula on immune function of asthmatic mice: the levels of SOD,CAT and T-AOC in the mice of Dendrobium officinale formula were all increased,and the activities of SOD and CAT in the middle dose group were significantly increased(p<0.01),which were 84.46% and 61.29%,respectively,and the activity of T-AOC was significantly increased(p<0.001),which was 111.79%.The results of gene expression analysis of immune cytokines showed that the gavaging group of Dendrobium officinale formula had the best dose effect,the expression levels of IL-5,NF-?B,transcription factor GATA-3 and ROR?t decreased by 4.59,2.16,2.01 and 0.59 respectively,compared with the model group,while the expression levels of IFN-? and T-bet increased by 0.45 and 0.12 respectively.The results showed that Dendrobium officinale formula could improve the immunity of mice by improving the antioxidant capacity of the asthma mice and participating in the metabolic pathways of Th1,Th2,Th17 and NF-?B to improve the inflammation of the mouse trachea.3.Effects of Dendrobium officinale formula on intestinal flora of asthmatic mice: total DNA was extracted from the intestinal contents of each group of mice,and sequencing analysis was conducted,so as to compare the intestinal flora of 6 groups of mice.The results showed that: the intestinal flora of the asthmatic mice was different from that of the gavaging group and the normal group.The obvious ones were Lactobacillus,Akkermansia,Turicibacter,Rikenella and Faecalibaculum.The asthma group had 17.29%percent higher Lactobacillus than the normal group.Akkermansi and Turicibacter were lower 3.36%,0.72% than normal group,gavaging group of intestinal flora diversity has an obvious relationship with dose,low dose group of the most close to normal intestinal flora community groups,compared with normal group,Turicibacter and Akkermansia ratio increased by 10.35% and 6.09% respectively.Clustering analysis shows that the model group is the same as the normal group,which may be because the recovery period of the model group is relatively long.The above results indicated that Dendrobium officinale formula could regulate the intestinal flora of mice,and its effect may exceed the effect of asthma on intestinal flora.
